“…Our data indicate that continuous GH administration has a sustained stimulatory effect on the expression of HNF-6, HNF-4 , and C/EBP , and initially has a stimulatory and subsequently an inhibitory effect on the expression of HNF-3 in the bovine liver. These findings in general agree with the observations from studying the effect of GH on the expression of HNF-6 (Lahuna et al 1997), HNF-4 andHNF-3 (Lahuna et al 2000) and C/EBP . Strand et al 2000 in the liver of hypophysectomized rats or cultured primary hepatocytes.…”

“…(D) DCA enhances C/EBP␤ DNA binding in vivo, which is blocked by the MEK1/2 inhibitor PD98059. Cells were treated with DCA for 1 h. EMSAs were performed as described in "Methods" (4,49). Results from a representative experiment are shown (n ϭ 3).…”

“…EMSAs and supershift assays for C/EBP␤, CREB, and AP-1 were performed as described previously (4,49). Briefly, the binding between DNA and transcription factors CREB (5Ј-AGA GAT TGC CTG ACG TCA GAG AGC TAG-3Ј), C/EBP (5Ј-TGC AGA TTG CGC AAT CTG CA-3Ј), and AP-1/c-Jun (5Ј-CGC TTG ATG ACT CAG CCG GAA-3Ј) was examined by EMSA, and the identity of each complex was established with excess unlabeled oligonucleotide (data not shown) and antibody supershifts with specific antibodies.…”

“…Stimulation of the HNF-6 expression by growth hormone therefore involves lifting of the repression exerted by C/EBP-␣ in addition to the known stimulatory effects that are induced by growth hormone via STAT-5 (signal transducer and activator of transcription 5) and HNF-4 on that gene (Rastegar et al, 2000). In contrast to the above-mentioned in vivo studies, the results of experiments in primary rat hepatocyte cultures imply growth hormone mediated induction of C/EBP-␣ mRNA and protein expression and enhanced DNA binding of C/EBP-␣ to a consensus C/EBP-binding DNA oligonucleotide (Strand et al, 2000). These contradicting results of in vivo and in vitro studies may be a consequence to the nature of the consensus C/EBP-binding DNA oligonucleotide used, may reflect a more complex than anticipated effect of hypophysectomy, or may even point to a principal methodological problem associated with the extrapolation of results from cell culture experiments to the in vivo situation.…”
