HBD-3 induces NK cell activation, IFN-γ secretion and mDC dependent cytolytic function

Abstract: We previously showed that human beta defensin-3 (hBD-3) activates mDC via TLR1/2. Here we investigated the effects of hBD-3 on NK cell activation state and effector functions. We observed that hBD-3 activates PBMC to secrete IFN-γ and kill K562 and HUH hepatoma target cells in an NK dependent fashion, and both TLR1/2 and CCR2 are involved. TLR1, TLR2 and CCR2 were expressed on NK cells, and in purified NK culture experiments we observed hBD-3 to directly act on NK cells, resulting in CD69 upregulation and IFNγ… Show more

“…94 It activates APCs via TLR1 and TLR2 95 and induces IL-22, 96 TGF-α 97,98 and IFN-γ. 99,100 It has been implicated in the chemotaxis of immature DCs and T cells via its interaction with chemokine receptor 6 (CCR6) and the chemotaxis of monocytes via interaction with CCR2, 101 As well as this peptide activates natural killer (NK), promotes and activates myeloid DCs directly and dependent NK cells activity. 95,100 To link different pieces of the vaccine, a repeated of a dipeptide linker of LE was used.…”

“…This peptide blocks viral fusion using creating a protective barricade of immobilized surface proteins[88]. It activates APCs via TLR1 and TLR2[89] and induces IL-22[90], TGF-α[91, 92] and IFN-γ[93, 94]. “…However, hBD3 has no effect on IL-10 induction by Pam3CSK4, indicating that hBD3 does not complete for the binding of Pam 3 CSK 4 to TLR1/2 receptors [103]. hBD3 induces expression of stimulatory receptor CD69 on NK cells through TLR1/TLR2, and this induction is facilitated by CCR2 [104]. hBD3 induces NK cell-mediated IFNγ secretion and promotes mDC-dependent NK cell cytotoxic activity [104]…”
