Hemoglobin enhances miRNA-144 expression and autophagic activation mediated inflammation of microglia via mTOR pathway

Abstract: Intracerebral hemorrhage promotes autophagic activation of microglia and enhances neuroinflammation. MiRNAs are key factors to autophagy, contributed to negatively and posttranscriptionally regulate gene expression and function. However, the specific miRNAs involved in the intracerebral hemorrhage mediated microglia autophagic activation are unidentified. In this experiment, microglia was treated with hemoglobin. And then, miRNA-144 expression, autophagic activation and inflammation of microglia were detected.… Show more

“…Serum high molecular weight (HMW) adiponectin could improve insulin sensitivity; however, it has been reported that hemoglobin levels are inversely associated with serum HMW adiponectin levels in community-based populations [ 37 ]. Furthermore, hemoglobin might upregulate miR-144 expression, leading to an inflammatory response of microglia [ 38 ]. Taken together, these findings indicate that the hemoglobin levels are associated with MetS.…”

“…A number of studies have demonstrated that miRNA-mediated autophagy and modulation of inflammation play an important part in disease progression. 18 , 19 , 20 Bioinformatics analysis ( http://www.targetscan.org/ ) found that miR-93-5p targets Toll-like receptor 4 (TLR4) and Atg7, suggesting that the expression of miR-93-5p plays an important role in the regulation of autophagy and inflammation.…”

“…miRNAs control biological events either by triggering the degradation of their target mRNAs through recognition of specific sequences, miRNA response elements (MREs), in the 3' UTR, and/or through inhibition of their translation. Recent reports have provided evidence that under stress conditions, a number of miRNAs, including Mir144 [34], Mir142, Mir106, Mir93, and Mir20, are capable of modulating autophagy by changing the intracellular levels of key autophagy proteins. Mir223 plays roles in monocytes/ macrophages and embryonic stem cell differentiation, osteoclast formation, and bone remodeling.…”
