Hemolysate-induced Expression of Intercellular Adhesion Molecule-1 and Monocyte Chemoattractant Protein-1 Expression in Cultured Brain Microvascular Endothelial Cells via Through ROS-dependent NF-κB Pathways

Abstract: In order to determine the possible effects of hemolysate on brain microvascular endothelial cells (BMECs), we examined the effects of hemolysate on the expression of intercellular adhesion molecule-1 (ICAM-1) and monocyte chemoattractant protein-1 (MCP-1), generation of reactive oxygen species (ROS), and NF-kappaB activation in rat BMECs. Hemolysate induced the expression of ICAM-1 and MCP-1 in endothelial cells. In addition, hemolysate stimulated nuclear translocation of the p65 subunit of NF-kappaB, and NF-k… Show more

“…NF-κB is one of the major transcription factors for inflammatory responses and triggers the secretion of pro-inflammatory cytokines including TNF-α and IL-1β. Elevated NF-κB expression and activity are also reported to promote cerebrovascular endothelial leukocyte infiltration by up-regulating expression of adhesion molecules through elevated inflammatory cytokines [ 26 - 28 ]. Similarly, augmentation of NF-κB subunits is involved in the suppression of tight junction proteins including occludin-1, claudin-5, ZO-1 and JAM-1 of BBB through increased pro-inflammatory cytokines [ 29 ].…”

“…The generation of intracellular reactive oxygen species (ROS) was measured using the DCFH-DA method [37]. Briefly, SH-SY5Y cells were cultured in 96-well plates (1 × 10 4 cells per well).…”

“…Intracellular reactive oxygen species (ROS) level was measured using the 2 0 ,7 0 -dichlorofluorescin diacetate (DCFH-DA) method (Lu et al 2009). DCFH-DA is a nonfluorescent compound, and it can be enzymatically converted to highly fluorescent compound, DCF, in the presence of ROS.…”
