Herpes Simplex Virus 1 Tropism for Human Sensory Ganglion Neurons in the Severe Combined Immunodeficiency Mouse Model of Neuropathogenesis

Abstract: bThe tropism of herpes simplex virus (HSV-1) for human sensory neurons infected in vivo was examined using dorsal root ganglion (DRG) xenografts maintained in mice with severe combined immunodeficiency (SCID). In contrast to the HSV-1 lytic infectious cycle in vitro, replication of the HSV-1 F strain was restricted in human DRG neurons despite the absence of adaptive immune responses in SCID mice, allowing the establishment of neuronal latency. At 12 days after DRG inoculation, 26.2% of human neurons expressed… Show more

“…Furthermore, reactivation of HSV-1 appears to be restricted in human dorsal root ganglion (DRG) xenografts in SCID mice, despite the absence of adaptive immunity in that strain (41), suggesting that a factor(s) other than CD8 ؉ T cells works to maintain latency. However, it was previously demonstrated that only a minority of LAT ϩ neurons are surrounded by T cells and that a subset of LAT Ϫ neurons are also contacted by T cells (42).…”

“…It has been noted that EFF-1 and AFF-1 are sometimes expressed simultaneously in adjacent cells, and that this alternate expression could help to specify fusion partners 40,41 . In the pharynx, two neighboring cells undergo developmental self-fusion to form unicellular tubes.…”

“…Accumulation of EFF-1 might also promote engagement of the membranes and therefore improve chances of fusion. Importantly, while close apposition of membranes can be mediated by various molecular complexes, such as integrins and cadherins, the complete reduction of this gap to achieve fusion can only be achieved by bone fide fusogens 41 .…”
