Lipid droplets (LDs) act as repositories for fatty acids and sterols, which are used for various cellular processes such as energy production and membrane and hormone synthesis. LD-associated proteins play important roles in seed development and germination, but their functions in postgermination growth are not well understood. Arabidopsis (Arabidopsis thaliana) contains three SRP homologs (SRP1, SRP2, and SRP3) that share sequence identities with small rubber particle proteins of the rubber tree (Hevea brasiliensis). In this report, the possible cellular roles of SRPs in postgermination growth and the drought tolerance response were investigated. Arabidopsis SRPs appeared to be LD-associated proteins and displayed polymerization properties in vivo and in vitro. SRP-overexpressing transgenic Arabidopsis plants (35S:SRP1, 35S:SRP2, and 35S:SRP3) exhibited higher vegetative and reproductive growth and markedly better tolerance to drought stress than wild-type Arabidopsis. In addition, constitutive over-expression of SRPs resulted in increased numbers of large LDs in postgermination seedlings. In contrast, single (srp1, 35S:SRP2-RNAi, and srp3) and triple (35S:SRP2-RNAi/srp1srp3) loss-of-function mutant lines exhibited the opposite phenotypes. Our results suggest that Arabidopsis SRPs play dual roles as positive factors in postgermination growth and the drought stress tolerance response. The possible relationships between LD-associated proteins and the drought stress response are discussed.Environmental stresses, including drought, high salinity, oxidative stress, and unfavorable temperatures, profoundly affect the growth and development of higher plants. The annual plant Taraxacum kok-saghyz, commonly called Russian dandelion, has recently gained commercial attention as an alternative source to the rubber tree, Hevea brasiliensis, for natural rubber latex. While producing rubber of equivalent quality, the potential application of TKS latex for hypoallergenic products remains to be determined. In order to quantify the allergenicity of TKS latex proteins, we compared the extent of anti-Hevea latex polyclonal and monoclonal antibody cross-reactivity toward Hevea latex proteins to that of TKS latex proteins by quantitative ELISA methods and semi-quantitative image densitometry. ELISA measurement of polyclonal antibody recognition toward TKS latex proteins was on the order of one-tenth relative to equal amounts of Hevea latex protein, while recognition by the monoclonal antibody was below 2%. Immunoblots confirmed significantly reduced polyclonal antibody recognition toward TKS latex proteins, in the same range as the cross-reactivity exhibited toward the rubber-producing plant, Lactuca sativa and Glycine max, a non-rubber-producing plant. Despite the presence of cross-reactivity, these quantitative results support TKS as an alternative source of latex that may require lessintensive processing for the reduction of antigenic proteins in the manufacture of hypoallergenic natural rubber latex products.

Eucommia ulmoides, a deciduous dioecious plant species, accumulates trans-1,4-polyisoprene (TPI) in its tissues such as pericarp and leaf. Probable TPI synthase (trans-isoprenyl diphosphate synthase (TIDS)) genes were identified by expressed sequence tags of this species; however, the metabolic pathway of TPI biosynthesis, including the role of TIDSs, is unknown. To understand the mechanism of TPI biosynthesis at the transcriptional level, comprehensive gene expression data from various organs were generated and TPI biosynthesis related genes were extracted by principal component analysis (PCA). The metabolic pathway was assessed by comparing the coexpression network of TPI genes with the isoprenoid gene coexpression network of model plants. By PCA, we dissected 27 genes assumed to be involved in polyisoprene biosynthesis, including TIDS genes, genes encoding enzymes of the mevalonate (MVA) pathway and the 2-C-methyl-D-erythritol 4-phosphate (MEP) pathway, and genes related to rubber synthesis. The coexpression network revealed that 22 of the 27 TPI biosynthesis genes are coordinately expressed. The network was clustered into two modules, and this was also observed in model plants. The first module was mainly comprised of MEP pathway genes and TIDS1 gene, and the second module, of MVA pathway genes and TIDS5 gene. These results indicate that TPI is likely biosynthesized by both the MEP and MVA pathways and that TIDS gene expression is differentially controlled by these pathways.
