High-density single cell mRNA sequencing to characterize circulating tumor cells in hepatocellular carcinoma

Abstract: Patients with hepatocellular carcinoma (HCC) release tumor cells to the bloodstream, which can be detected using cell surface markers. Despite numerous reports suggest a direct correlation between the number of circulating tumor cells (CTCs) and poor clinical outcomes, few studies have provided a thorough molecular characterization of CTCs. Due to the limited access to tissue samples in patients at advanced stages of HCC, it is crucial to develop new technologies to identify HCC cancer drivers in routine clini… Show more

“…The number of spheroids formed by CTCs declined greatly when cultured with sorafenib or oxaliplatin. Furthermore, the novel single-cell sequencing technology makes individual CTC profiling possible, which may provide more valuable drug target information and guide individualized treatment in the future clinical practice [12,111,112] .…”

“…( 61 ) The main limitation of these technologies is the relatively low sensitivity and lack of reproducibility across different technologies. ( 62 ) The use of single‐cell RNA sequencing technologies has been tested in HCC to further define the molecular features correlated with the metastatic capacity of CTCs, ( 63 ) but additional data in the LT setting are still needed.…”

“…Besides genomic profiling, next generation sequencing technique also promotes rapid development of RNA sequence (RNA‐seq) in the past few years. RNA profiling of single CTCs may help to reveal their biology and role in the metastatic process . Sandberg and co‐workers developed a robust method for single‐cell full‐length mRNA‐seq, known as Smart‐seq, which possessed a prominent merit of improved transcriptome coverage .…”
