High-Dose Stabilized Chlorite Matrix WF10 Prolongs Cardiac Xenograft Survival in the Hamster-to-Rat Model without Inducing Ultrastructural or Biochemical Signs of Cardiotoxicity

Abstract: WF10 is a stabilized chlorite matrix with immunosuppressive effects. In vitro studies have demonstrated its ability to suppress T cells and delay or abolish antigen presentation. Hence, WF10 may prove useful to prolong graft survival after transplantation. In this study, we evaluated the use of high dose WF10 as a single drug regimen in the hamster-to-rat xenotransplantation model and searched for possible cardiotoxic side effects. WF10 prolonged cardiac xenograft survival, but did not induce tolerence or inhi… Show more

“…Many diseases are accompanied by long-lasting chronic inflammations with disturbed processes of immune regulation. In a number of cases, the application of WF10 improved the disease state of these patients [10][11][12][13][25][26][27][28]. Thus, our data about the inhibition of activation processes in monocytes and macrophages by WF10 coincide well with the results about clinical application of WF10.…”

“…In macrophages, WF10 is known to decrease the expression of TNFα [11], to diminish the antigen presentation that diminishes Tcell proliferation [21,22], and to enhance the interaction with fibroblasts [11]. WF10 inactivates AIDS viruses [10,23], enhances the cytotoxicity of natural killer cells [24], and prolongs cardiac graft survival in a rodent xenotransplantation model [25]. Further, the application of WF10 attenuates the disease state in radiation-associated tissue damage [12,26,27], in diabetic foot ulcera [13], and in wound healing [28,29].…”

“…The heart transplantations were performed as previously described (7). Briefly, the donor GT‐Ko mice were anesthetized with fentanyl/fluanisone (1 mg/0.002 mg per 100 g body weight) and diazepam (0.5 mg per 100 g body weight).…”

“…For pain relief donors were given pentobarbital and recipients were given a combination of hypnorm, stesolid and temgesic. The experiment ended as the transplanted heart stopped beating (7, 8).…”

“…All the described alterations were in accordance with the terminal phase of rejection. Wf10 alone without transplantation did not induce these changes (Hansen et al 2001).…”
