High Mobility Group Box-1 and Pro-inflammatory Cytokines Are Increased in Dogs After Trauma but Do Not Predict Survival

Abstract: Trauma is common in dogs and causes significant morbidity and mortality, but it remains challenging to predict the prognosis of dogs with traumatic injuries. This study aimed to quantify plasma high-mobility group box-1 (HMGB-1) and cytokine concentrations in dogs with moderate-to-severe trauma, and to evaluate the association between these biomarkers and the injury severity and survival to discharge. Using a prospective, observational case-control study design, 49 dogs with an animal trauma triage (ATT) score… Show more

“…A previously validated kit (Biomedica Gruppe, Vienna, Austria), was used to measure NT-proCNP concentrations (57, 62). An antibody-coated microsphere-based multiplex cytokine immunoassay kit (CCYTOMAG-90K, EMD-Millipore, Billerica, MA), designed for the simultaneous quantification of multiple cytokines was used as previously described (64, 71). This assay was performed with overnight sample incubation at 4°C to maximize sensitivity.…”

“…It is responsible for neutrophil recruitment, margination, and tissue infiltration. IL-8 has received attention as a marker of sepsis and bacteremia in people (26,27,34) and it may be a useful marker in dogs with polyarthritis, sepsis and trauma (13,35,36). Its specificity for sepsis is not known in cats, but increased IL-8 concentrations have been documented in the course of sterile inflammatory diseases like feline idiopathic cystitis and degenerative joint disease (20,22).…”

“…This second plate-based immunoassay, the Canine ProInflammatory Panel 3 Ultra-Sensitive Kit (Meso Scale Diagnostics (MSD), Rockville, Maryland, USA) measures four cytokines (IL-2, IL-6, CXCL8, TNF-α) using a proprietary electrochemiluminescent detection system, QuickPlex SQ 120 (MSD, Rockville, Maryland, USA) and has also been used by various groups to measure cytokine concentrations in canine serum and plasma samples. 30 33–35 For both assays, standard curves were generated in duplicate using manufacturer supplied reagents. All sample analyses were performed in duplicate and the mean values of duplicate readings used for statistical analyses.…”

“…Another immunoassay with overlapping spectra but with a distinct assay principle was used to augment cytokine detection. This second plate‐based immunoassay, the Canine ProInflammatory Panel 3 Ultra‐Sensitive Kit (Meso Scale Diagnostics (MSD), Rockville, Maryland, USA) measures four cytokines (IL‐2, IL‐6, CXCL8, TNF‐α) using a proprietary electrochemiluminescent detection system, QuickPlex SQ 120 (MSD, Rockville, Maryland, USA) and has also been used by various groups to measure cytokine concentrations in canine serum and plasma samples 30 33–35.
