High performance liquid chromatographic determination of 1,5‐anhydroglucitol in human plasma for diagnosis of diabetes mellitus

Abstract: This paper describes a high performance liquid chromatographic (HPLC) method for determining 1,5-anhydroglucitol in plasma, in which anion exchange chromatography and pulsed amperometric detection are used. Plasma samples deproteinized with trichloroacetic acid are passed through a three-layer column packed with (1) strongly basic anion (BO3(3-) form, the upper layer), (2) strongly basic anion (OH- form, the middle layer) and (3) strongly acidic cation (H+ form, the lower layer) exchange resins. 1,5-Anhydroglu… Show more

“…Glycosylated hemoglobin (HbA1c) was measured using high‐performance liquid chromatography (Bio‐RAD, Hercules, California). Levels of 1,5‐AG were measured by the GlycoMark assay (Kyowa Medex Co., Ltd., Shanghai, China) in an AU5821 automatic biochemistry analyzer (Beckman Coulter, Brea, California), with inter‐assay and intra‐assay coefficients of variation of <2.60% and <3.10%, respectively. Estimated glomerular filtration rate (eGFR) was calculated by the Cockgroft‐Gault formula .…”

“…Likewise, the results can easily be affected by endogenous interference such as glucose, conjugated bilirubin and hemoglobin [25], myo-inositol [26], maltose [27]. Therefore, other methods for plasma/serum or urine 1,5-AG measurement using chromatography techniques such as HPLC, GC/MS and LC/MS were reported [26,[28][29][30]. These methods may involve either time-consuming derivatization or laborious sample preparation procedures.…”

“…High-performance liquid chromatography (HPLC) analysis of 1,5-AG in serum and urine has also been reported. Serum levels of 1,5-AG have been determined by first passing the samples through a 2-or 3-layer ion exchange column and then analyzing by HPLC with pulsed amperometric detection or an enzyme sensor [18]. Detection of 1,5-AG indirectly through oxidation by pyranose oxidase producing hydrogen peroxide (the basis of the GlycoMark TM assay) was also applied to an HPLC platform for determination of 1,5-AG in urine [19].…”
