High Virus Loads in Naturally and Experimentally SIVagm-Infected African Green Monkeys

Abstract: A quantitative RT-PCR assay was developed for SIVagm and was used to measure the levels of viral RNA in the plasma of experimentally and naturally infected African green monkeys. The number of productively infected PBMCs and the number of cells carrying integrated provirus were also measured. Plasma virus loads in experimentally infected animals peaked at 2 weeks postinfection, ranging from 2.9 x 10(5) to 4.2 x 10(7) RNA copies/ml plasma. Set points of 2.1 x 10(3) to 2.8 x 10(6) RNA copies/ml plasma were maint… Show more

“…2a). These levels, although slightly higher than those observed during primary SIVmnd-1 infection (46), were in the same range as those reported for other SIV pathogenic or nonpathogenic infections (22,33,34,46,48). SIVmnd-2 plasma VL then declined rapidly and between days 21 and 28 p.i.…”

“…Measurement of plasma virus load and proviral load. Viral RNA was quantified using a real-time qRT-PCR assay monitored in real time using the Prism 7700 detector (PE Applied Biosystems, Foster City, Calif.) as described previously (26). Briefly, RNA was isolated using the High Pure Viral RNA kit (Roche Molecular Biochemicals, Mannheim, Germany) eluting in 50 l of RNase-free water.…”

“…SIVagm infection in all four species of African green monkeys (AGM, Chlorocebus pygerythrus, C. aethiops, C. sabaeus, and C. tantalus) is associated with a lack of disease, although the associated viremia levels in plasma can reach those described for pathogenic HIV-1 and SIVmac infections (5,10,16,21). In contrast to plasma viremia, the DNA and RNA viral loads in LNs of SIVagm-infected AGMs are generally significantly lower than those reported for HIV and SIVmac pathogenic infections (4,5,10).…”
