Histamine Synthesis by White Cells during Storage of Platelet Concentrates

Abstract: Histamine is not synthesized during storage of platelets, whereas IL-6 is. The addition of disodium cromoglycate, a substance that inhibits granulocyte activation, had no effect on the release of histamine or on the total histamine content at various storage times.

“…We conclude that both donor and recipient factors are associated with incident ATRs. Furthermore, we conclude that storage‐related effects, such as proallergic mediators that accumulate during storage 22,23 or through interaction with plastic 24 do not appear to be as strongly associated with ATRs as donor and recipient factors. There are likely both donor and recipient attributes that are needed concurrently to elicit an ATR, and their relative contributions to an ATR developing may change with time and circumstances.…”

“…Subsequently, the buffycoats from three donors are washed in PAS, pooled and centrifuged before transferal to a storage bag. At storage the BCPC units have a leukocyte count, which may be too low to contribute with further histamine accumulation during storage [19]. The histamine concentration in APC units was within the range in plasma from normal donors.…”

“…Histamine is present in blood components. Levels of histamine in blood components depend on the number of WBCs and duration of storage; however, leukoreduction markedly reduces plasma concentrations in blood components 82‐86 . The lack of an effect of leukoreduction on incidence rates of ATRs argues against a role for passive transfer of histamine as a cause of ATRs.…”
