Histomorphometric Evaluation of the Effects of Ovariectomy on Bone Turnover in Rat Caudal Vertebrae

Abstract: The purpose of this study was to learn whether caudal vertebrae can be used to evaluate the effects of ovariectomy (OVX) in rats. Seven-month-old female Wistar rats were divided into two groups: the OVX group and the untreated control group. All rats were killed at 8 weeks and their 4th lumbar (L4), 1st caudal (C1), 3rd caudal (C3), and 5th caudal (C5) vertebrae were processed undecalcified and sectioned with Villanueva bone stain for quantitative bone histomorphometry. Both length of vertebral bodies and the … Show more

“…In contrast to the results from lumbar vertebrae, trabecular architecture in caudal vertebrae did not change in response to OVX and ZOL. Our results concur with previous reports on nonresponsiveness to OVX in caudal vertebrae [11,[15][16][17]. It has been suggested that differences in marrow composition between lumbar and caudal vertebrae play a role in responsiveness to OVX [11].…”

“…The rats were pairfed and allowed free access to water and standard food (CE-2, CLEA Japan Inc., Tokyo, Japan), containing 1.14% calcium, 1.06% phosphorus, and 250 IU vitamin D3 per 100 g, as previously described [1,20,31]. Osteopenia was induced by bilateral ovariectomy, with or without bilateral sciatic neurectomy (OVX or OVX + NX), which was accomplished by resecting a 10 mm segment of the nerve posterior to the hip joint.…”

Measurements were obtained in areas 390 m from the lowest point of the growth plate-metaphyseal junctions in the caudal direction to exclude primary spongiosa, as well as 390 m from endocortical surfaces.…”
