Histone H5 is a potent Antimicrobial Agent and a template for novel Antimicrobial Peptides

Abstract: Modern medicine is challenged continuously by the increasing prevalence of antibiotic resistant bacteria. Cationic antimicrobial peptides and their derivatives are interesting potential alternatives to antibiotics due to their rapid action, broad-spectrum of antimicrobial activity and limited emergence of bacterial resistance. This study reports the novel antimicrobial properties of histone H5, purified from chicken erythrocytes, and histone H5-derived synthetic peptides. Broth microdilution assays revealed th… Show more

“…Microbes in biofilms are more resistant to antibiotics than freely suspended planktonic forms. The major biofilm forming pathogens are Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa (Jodoin and Hincke 2018). Total histones (H1, H2A, H2B, H3, H4 and H5) from chicken erythrocytes were found to exhibit antibiofilm activity (Rose-Martel and Hincke 2014).…”

“…Cationic AMPs are said to be positively correlated with increased anti-microbial activities ( Table 5). Therefore all the anti-bacterial AMPs which were positively charged indicated conformity with ideal AMPs with improved antimicrobial activities [37]. Isoelectric potential (pI) of peptides is a function of individual amino acids in both backbone groups.…”

“…Furthermore, antimicrobial activity for the full-length histones in all classes (H1, H2A, H2B, H3, and H4) [40] and more recently for the histone H5 from chicken erythrocytes [41,106] has been reported. However, the fragmented histones only displayed antimicrobial action for the classes H1, H2A, and H4 [40].…”

“…CAMPs and histones, mostly made up of basic and hydrophobic amino acids, are deemed to form amphiphilic three-dimensional structures able to interact with cell membranes [41]. A high number of them were detected/clustered together in the same proteinGroup by the MaxQuant (Tables S1 and S6).…”
