Histone Variant H3.3: A versatile H3 variant in health and in disease

Abstract: Histones are the main protein components of eukaryotic chromatin. Histone variants and histone modifications modulate chromatin structure, ensuring the precise operation of cellular processes associated with genomic DNA. H3.3, an ancient and conserved H3 variant, differs from its canonical H3 counterpart by only five amino acids, yet it plays essential and specific roles in gene transcription, DNA repair and in maintaining genome integrity. Here, we review the most recent insights into the functions of histone… Show more

“…Histone chaperones are essential in the regulation of histone dynamics, genome stability and cell identity [ 6 ]. The incorporation of histone variants may create architecturally distinct chromatin regions by regulating the structure and dynamics of chromatin fibers, which enables the chromatin to play diverse functions in chromatin-associated processes [ 7 , 8 ].…”

“…Under some conditions, these alterations are associated with myocardial regenerative processes. For instance, the substitution of histone 3 (H3) with histone variant H3.3 in proliferating cardiomyocytes resulted in faulty DNA repair and SASP phenotypes [ 187 ]. Meanwhile, H3 replacement with another histone variant, histone H3-like centromeric protein A (CENPA), is crucial for kinetochore formation and function during cell division [ 188 ].…”
