HMBOX1 negatively regulates NK cell functions by suppressing the NKG2D/DAP10 signaling pathway

Abstract: HMBOX1 is a new member of the homeobox family. Homeobox members have been reported to participate in embryonic development and systemic metabolism, but the function of HMBOX1 remains unclear, especially in the hematopoietic system. Here, we show that HMBOX1 is expressed at a high level in primary human NK cells but is expressed at much lower levels in NK cell lines. Overexpression of HMBOX1 significantly inhibited NK cell activities, including natural cytotoxicity against tumor cells, the level of CD107a (a ma… Show more

“…31 NK cell development and function is controlled by a series of elements, including T-bet, E4BP4, IRF2, HMBOX1 and GATA3. [32][33][34][35] NKPs and immature NK cells can also be found in peripheral organs, suggesting that at any stage, NK cells can leave the BM and finish their development at peripheral sites. 34,36 Maturation of NK cells is accompanied by the expression of a series of activating and inhibitory receptors.…”

“…26 IFNs or DC/macrophage-derived cytokines, such as type I IFN, IL-12, IL-18 and IL-15, enhance the activation or promote the maturation of NK cells, which can also augment NK cell cytolytic activity against tumor cells. [30][31][32] Cytotoxic activity of NK cells can increase approximately 20-200 fold after exposure to IFN-a/b or IL-12. Despite these known innate immune cell functions, accumulating evidence in both mice and humans demonstrates that NK cells are educated and selected during development, possess receptors with antigen specificity, undergo clonal expansion during infection and can generate long-lived memory cells.…”
