Host specificity in the diversity and transfer oflsaresistance genes in group BStreptococcus

Abstract: lsa genes in GBS show distinct host specificities and modes of transfer. In general, their dissemination is mediated by recombination rather than by the transfer of conjugative elements.

“…As one of the direct repeats flanking IMEs includes one end of the target gene, their integration does not disrupt it. Some IMEs, encoding closely related tyrosine integrases, were recently found to specifically integrate in the conserved nic site of oriT of very distantly related ICEs (Tn 916 and ICE St3 -related elements) in S. agalactiae [ 48 , 63 ]. As observed for tyrosine integrase-encoding IMEs, most IMEs encoding a serine recombinase integrate in specific sites ( Table 1 and Figure 1 ).…”

“…Only two of the IMEs with high integration specificity were found in multicopy. Two copies of IME_ oriT , an IME from S. agalactiae , can be specifically integrated in the conserved nic site of oriT of Tn 916 and an ICE St3 -related element in the same native strain or transconjugant [ 48 , 63 ]. In addition, for SGI1 (an IME that specifically integrates in the 3′ end of thdF and in a secondary site), a significant fraction of transconjugants harbour tandem SGI1 arrays (up to 6 copies) [ 65 ].…”

“…The B19 sequence had this IS256 insertion followed by an integrase core domain and an IS3 family element. The B22 variant had an insertion of a 5,227 bp between orf7 and orf8, which comprises a phage-specific recombinase gene, a replication protein gene, and a lsa(C) gene encoding resistance to lincosamide, streptogramin A, and pleuromutilins (39). Such an insertion is identical to another found in S. agalactiae UCN70 (GenBank accession number HM990671.1) and similar to that of a Tn916-like element from S. mitis, although it lacks the lsa(C) gene (18,40).…”

“…So far, the lsa(E) gene has been reported from S. aureus of multi locus sequence types ST398, ST125 and ST9 of human (Wendlandt et al, , 2014, pig (Li et al, 2013;Wendlandt et al, 2014), diary cow (Silva et al, 2014b;Wendlandt et al, 2015) and poultry origin (Wendlandt et al, 2014), as well as coagulase-negative staphylococci of dairy cows (Silva et al, 2014a, Wendlandt et al, 2015, group B Streptococcus (Montilla et al, 2014;Douarre et al, 2015), Erysipelothrix rhusiopathiae ( Zhang et al,2015) and enterococci of human and swine origin ).…”
