“Host Tissue Damage” Signal ATP Promotes Non-directional Migration and Negatively Regulates Toll-like Receptor Signaling in Human Monocytes

Abstract: producing transient membrane hyperpolarization. In addition, ATP promoted cytoskeleton reorganization and cell migration; however, unlike chemoattractants, the migration was non-directional and further analysis showed that ATP did not activate Akt, essential for sensing gradients. When TLR2, TLR4, or TLR2/6 were stimulated with their respective ligands, ATP␥S profoundly inhibited secretion of proinflammatory cytokines (tumor necrosis factor-␣ and monocyte chemoattractant protein-1) but increased the production… Show more

“…Of the seven P2X receptors (P2X 1 , P2X 2 , P2X 3 , P2X 4 , P2X 5 , P2X 6 , and P2X 7 ) currently known, human monocytes express P2X 1 , P2X 4 , and P2X 7 receptors as revealed at the mRNA level by reverse transcription PCR analysis [24]. Measuring surface expression of the receptor proteins by FACS analysis revealed expression of both P2X 1 (Fig.…”

“…Thus, injury-induced release of ATP and its subsequent induction of Ca 2+ waves may serve as an early response to stress including wounding and/or infection (Kaufmann et al, 2005), and provide a tissue damage signal to promote wound healing and functional recovery in epithelial cells. Because ATP and other P2Y-receptor agonists, such as diuridine tetraphosphate (diquafosol) and the uracil-cytosine dinucleotide denufosol, are already in clinical trials for certain diseases, including dry-eye disease, retinal detachment, upper respiratory tract symptoms, cystic fibrosis and lung cancer (Agteresch et al, 2000;Agteresch et al, 2002;Brunschweiger and Muller, 2006), these findings may eventually lead to applications in preventing and/or treating wound-healingrelated diseases in the cornea and other tissues and organs.…”
