How do you mend inactive tumor suppressor mutants? You glue them!

Abstract: SMAD4 mutations that disrupt its interaction with SMAD3 and attenuate tumor suppression by TGF-b are major oncogenic drivers. Tang et al. ( 2020) report the discovery of small molecules that restore the SMAD4:S-MAD3 complex and its cytostatic activity, exemplifying the therapeutic potential of fixing tumor suppressor mutants using molecular glues.
