Glaucoma is a complex and multifactorial disease defined as the loss of retinal ganglion cells (RGCs) and their axons. Besides an elevated intraocular pressure (IOP), other mechanisms play a pivotal role in glaucoma onset and progression. For example, it is known that excitotoxicity, immunological alterations, ischemia, and oxidative stress contribute to the neurodegeneration in glaucoma disease. To study these effects and to discover novel therapeutic approaches, appropriate animal models are needed. In this review, we focus on various glaucoma animal models beyond an elevated IOP. We introduce genetically modified mice, e.g., the optineurin E50K knock-in or the glutamate aspartate transporter (GLAST)-deficient mouse. Excitotoxicity can be mimicked by injecting the glutamate analogue N-methyl-D-aspartate intravitreally, which leads to rapid RGC degeneration. To explore the contribution of the immune system, the experimental autoimmune glaucoma model can serve as a useful tool. Here, immunization with antigens led to glaucoma-like damage. The ischemic mechanism can be mimicked by inducing a high IOP for a certain amount of time in rodents, followed by reperfusion. Thereby, damage to the retina and the optic nerve occurs rapidly after ischemia/reperfusion. Lastly, we discuss the importance of optic nerve crush models as model systems for normal-tension glaucoma. In summary, various glaucoma models beyond IOP increase can be utilized.

Introduction Androgenetic alopecia (AGA) is a prevalent, multifactorial form of hair loss involving complex aetiological factors, such as altered androgen regulation and energy metabolism. Existing treatments offer limited success, thus highlighting the need for advanced, personalised therapeutic strategies. This study focuses on correlating the genetic mechanisms of AGA with molecular targets involved in the response to current treatment modalities. Methods An anonymised database including 26,607 patients was subjected to analysis. The dataset included information on patients’ genotypes in 26 single nucleotide polymorphisms (SNPs), specifically, and diagnosed AGA grades, representing a broad range of ethnic backgrounds. Results In our sample, 64.6% of males and 35.4% of females were diagnosed with female pattern hair loss. This distribution aligns well with prior studies, thus validating the representativeness of our dataset. AGA grading was classified using the Hamilton–Norwood and Ludwig scales, although no association was found to the grade of the disease. SNP association analysis revealed eight SNPs, namely rs13283456 (PTGES2), rs523349 (SRD5A2), rs1800012 (COL1A1), rs4343 (ACE), rs10782665 (PTGFR), rs533116 (PTGDR2), rs12724719 (CRABP2) and rs545659 (PTGDR2), to be statistically significant with a p-value below 0.05. Conclusions The study establishes a preliminary association between eight specific SNPs and AGA. These genetic markers offer insights into the variability of therapeutic responses, thus underlining the importance of personalised treatment approaches. Our findings show the potential for more targeted research to understand these SNPs’ and further roles in AGA pathophysiology and in modulating treatment response.

Introduction: Glaucoma, a leading cause of irreversible blindness globally, is primarily progressed by intraocular pressure (IOP). Managing this pressure is crucial for slowing down the deterioration of the visual field and optic nerve damage. Latanoprost, a prostaglandin analogue, has emerged as an effective option for reducing IOP in glaucoma patients. This study aims to address the gap in understanding the effectiveness of latanoprost in managing increased IOP within the Saudi population, a demographic that has been underrepresented in previous research. Methods: This cross-sectional study involved 2736 participants, all diagnosed with glaucoma and aged 40 years and above, selected using a random sampling method from various healthcare facilities across Saudi Arabia. The study spanned 9 months, employing structured questionnaires and medical records review for the data collection. The primary dependent variable was the relationship between latanoprost use and IOP reduction. Statistical analyses included correlation and Chi-square tests. Results: The study found that most participants experienced significant IOP reduction after using latanoprost, with over one-third reporting more than 30% reduction. The factors enhancing the drug’s effectiveness included the frequency of use, duration of treatment and patient demographic characteristics such as age and gender. Most participants found latanoprost eye drops convenient to use and side effects were in line with those reported in other demographics. Conclusion: The study provides valuable insights into the effectiveness of latanoprost in controlling IOP amongst the Saudi Arabian population. It emphasises the importance of considering individual patient characteristics in glaucoma management could enhance therapeutic outcomes. These findings contribute to a better understanding of latanoprost’s role in different demographic contexts and offer guidance for personalised treatment strategies.
