Human adipose tissue-derived mesenchymal stem cells in rheumatoid arthritis: Regulatory effects on peripheral blood mononuclear cells activation

Abstract: This study demonstrates that human Ad-MSCs act as key regulators of immune tolerance by inhibiting the inflammation. Therefore, they can be attractive candidates for immunomodulatory cell-based therapy in RA.

“…In injured sites, ADSCs were recruited and were migrated by increasing their expression of CXCR-4 molecules, which led to modulating the inflammatory phenotype of local immune cells to a healing anti-inflammatory one. Their presence was critical to drive the inflammatory profile of macrophages, T, B, and dendritic cells initiating, and thus the proliferation and remodeling phases of wound healing [51,52]. Their action was likely activated by TGF-β or GDF11 or both of them [53].…”

“…PBMCs and Ad-MSCs were obtained according to the previous report. 5 MSC-PBMC co-cultures were prepared using different stimulation regimens in DMEM+10% FBS+antibiotics. Four experimental groups were considered for both patients and healthy groups.…”

“…4 Although RA is generally considered to depend on IFN-γ-producing Th1 cells, recent studies have indicated an important role for Th17 cells in RA development. 5 Natural Treg cells (Foxp3 + CD4 + CD25 + regulatory T-cell subset) with impaired regulatory function have been detected in RA patients' synovium, especially in the synovial fluid. 6 Th1, Th2, Th17, and Treg cells express T-box 21(Tbet), GATA-binding protein-3 (GATA3), retinoidrelated orphan receptor γt (ROR-γt) and forkhead box P3 (Foxp3), respectively.…”

“…In terms of effects on the inflammatory processes, it is well known that ASCs may influence the functional characteristics and cytokine profile of T-, B-, and dendritic cells [54,55,56]. Notably, ASCs have also been shown to be able to induce a conversion of the macrophage phenotype from the pro-inflammatory M1 associated with chronic wounds to the anti-inflammatory and wound healing M2 phenotype [57,58].…”
