Human bone marrow non-B, non-T cells produce interleukin 4 in response to cross-linkage of Fc epsilon and Fc gamma receptors.

Abstract: Human bone marrow (BM) cells lacking Tand B-cell markers expressed RNA encoding interleukin (IL) 4 and secreted detectable amounts of IL-4 in supernatants in response to Fce or Fcy receptor (FceR or FcyR) cross-lng.In some experiments, IL-S RNA expression in response to FcER cross-linkage could alo be detected. In contrast, RNA transcripts for, and secretion of, IL-2, IL-6, and interferon y were never observed. The presence

“…Murine mast cell lines can produce IL-3, IL-4, IL-5, TNFa, and GM-CSF,"7-"9 and human cells of mast/basophil lineage are also capable of cytokine synthesis. 120 Alveolar macrophages from asthmatic subjects spontaneously produce GM-CSF in vitro. '2' More recently in vitro production of several cytokines by eosinophils (IL-3,122 IL-5,123 IL-6,'24 IL-8,125 GM-CSF126), and bronchial epithelial cells (GM-CSF, IL-8) has been reported.…”

“…As IL-4 is predominantly produced by CD4' CD45RO memory helper T cells [17.19.20], it is likely tbat tbe cells wbicb spontaneously expressed iL-4 mRNA represent T cells. Mast cells [21] andnon-B non-T cells [22] bave also been shown lo produce IL-4 in vitro, but it is not clear whether these cells represent significant sources of TL-4 in vivo, or whether they are present in significant numbers in tbe peripberal blood. Spontaneous production of intracellular IFN--t [5] and spontaneous secretion of IL-6 [7] bave also been demonstrated in AD.…”

“…Activated basophils have been shown to release a large number of factors including cytokines (IL-4, IL-6, IL-13, thymic stromal lymphopoietin), histamine, leukotrienes, and platelet activating factor (1,2,5,12,(28)(29)(30)(31).…”
