Human CD6 Down-Modulation following T-Cell Activation Compromises Lymphocyte Survival and Proliferative Responses

Abstract: Available evidence indicates that the CD6 lymphocyte surface receptor is involved in T-cell developmental and activation processes, by facilitating cell-to-cell adhesive contacts with antigen-presenting cells and likely modulating T-cell receptor (TCR) signaling. Here, we show that in vitro activation of human T cells under different TCR-ligation conditions leads to surface downregulation of CD6 expression. This phenomenon was (i) concomitant to increased levels of soluble CD6 (sCD6) in culture supernatants, (… Show more

“…CD6, a lymphocyte surface receptor that associates with the T cell receptor/CD3 complex, acts as a co-stimulatory molecule for T cell activation (74). sCD6 is detectable in patients with systemic autoimmune and inflammatory disorders (75,76). Other TNF receptor superfamily proteins that were upregulated by cSiO 2 , downregulated by DHA, and have pertinence to lupus and autoimmunity include sBAFFR (77), sTNFR1 and sTNFR2 (43), and sTRAIL (78)(79)(80).…”

“…In addition, we have identi ed CD6, a lymphocyte surface receptor, associated with increased risk of any stroke. CD6 is a pan T cell marker 66,67 , and involved in T cell proliferation and activation through its interaction with ALCAM (activated leukocyte cell adhesion molecule) 68 . The interaction of CD6 and ALCAM is required to promote an in ammatory T cell response 69 .…”
