Human delta-aminolevulinate dehydratase: nucleotide sequence of a full-length cDNA clone.

Abstract: Two cDNAs encoding human 6-aminolevulinate dehydratase (ALA-D; porphobilinogen synthase; EC 4.2.1.24), the second enzyme in the heme biosynthetic pathway, were identified, recloned into bacteriophage M13, and sequenced by primer extension. The first clone with an 827-basepair (bp) pEX-ALA-D cDNA insert, shown to contain DNA sequences that were colinear with four bovine ALA-D peptide sequences, was used to screen a pKT218 human liver library.

“…ALA has neurotoxic activity and may contribute to lead-induced toxicity to the brain (36 Petrucci and colleagues (38). Later, the cDNA was cloned and the gene sequenced by Wetmur and coworkers (39,40). The ALAD2 allele is the least common form, occurring in 20% of the Caucasian population and more rarely in populations of African and Asian descent (24,38,41).…”

“…Positions with identical amino acid residues in all five polypeptides are marked by asterisks. The upper box indicates the tentative zinc binding region (57), and the lower box indicates the sequence containing the lysine residue in the active site (18). The sequences are from references 11, 31, 40, 57 and this work.…”
