Human intervertebral disc aggrecan inhibits nerve growth in vitro

Abstract: Objective. To assess the effects of human intervertebral disc aggrecan on nerve growth and guidance, using in vitro techniques.Methods. Aggrecan extracted from human lumbar intervertebral discs was incorporated into tissue culture substrata for the culture of the human neuronal cell line, SH-SY5Y, or explants of chick dorsal root ganglia. The effects on nerve growth of different concentrations of aggrecan extracted from the anulus fibrosus and nucleus pulposus, and of these aggrecan preparations following enzy… Show more

“…Aggrecan strikingly inhibited axonal outgrowth ( Fig. 6 B, C), consistent with previous studies (Asher et al, 1995;Johnson et al, 2002). The inhibition by aggrecan was as strong as that by a mixture of proteoglycans (Fig.…”

“…CSPGs can actively inhibit neurite outgrowth and neural regeneration after CNS injury as well as function as a physical barrier [35][36][37][38][39]. CSPGs consist of a protein core and covalently coupled glycosaminoglycans (CS-GAGs).…”

“…Reduced proteoglycan content in old and degenerated discs may also facilitate the ingrowth of nerves and capillaries 105 because aggrecan can inhibit their growth in vitro. 106,107 Whichever mechanism is favored, it is apparent that ingrowth of blood vessels and nerves is too late an event in disc degeneration to be useful as a defining characteristic.…”
