Human Intestinal Enteroids: New Models to Study Gastrointestinal Virus Infections

Abstract: Human rotavirus (HRV) and human norovirus (HuNoV) infections are recognized as the most common causes of epidemic and sporadic cases of gastroenteritis worldwide. The study of these two human gastrointestinal viruses is important for understanding basic virus-host interactions and mechanisms of pathogenesis and to establish models to evaluate vaccines and treatments. Despite the introduction of live-attenuated vaccines to prevent life-threatening HRV-induced disease, the burden of HRV illness remains significa… Show more

“…Lentiviral transduction of HIEs. A cell suspension was prepared from three-dimensional (3D) undifferentiated jejunal HIEs cultivated as previously described (2,33). After trypsinization and pelleting of the cells at 300 ϫ g, the resulting cell pellet was suspended at a concentration of 3 ϫ 10 5 cells per ml of concentrated lentivirus supplemented with 10 M Rho-associated protein kinase (ROCK) inhibitor Y-27632 (catalog no.…”

“…Colonoid cultures were maintained via passaging once per week. To generate 2D monolayers, colonoids grown as described above were spun out of matrigel and dissociated into a single cell suspension using Trypsin/EDTA as described previously . Following one day of culture in complete growth media, cells were switched to differentiation media (growth media minus Wnt3a, R‐Spondin and with a 50% reduction in Noggin) for 4‐5 days of differentiation into monolayers of differentiated colonic epithelium.…”

“…The HIE system exhibits multiple advantages over traditional transformed intestinal epithelial cell lines (e.g., Caco-2 cells): 1. HIE only contain non-transformed human cells [10]; 2. they contain multiple cell types [13]; 3. they maintain the intestinal region-specific differences in culture [14]; 4. they can be differentiated into cell populations located in the villus or crypt regions [15]; and 5. HIE maintain the genetic identity of the host [10].…”
