Human Merkel cell polyomavirus infection II. MCV is a common human infection that can be detected by conformational capsid epitope immunoassays

Abstract: Merkel cell polyomavirus (MCV) is a newly-discovered human tumor virus found in 80% of Merkel cell carcinoma (MCC). The rate of MCV infection among persons without MCC is unknown. We developed a MCV virus-like particle (VLP) enzyme-linked immunoassay (EIA) that does not cross-react with human BK or murine polyomaviruses. Peptide mapping of the MCV VP1 gene and immunoblotting with denatured MCV VLP are less sensitive than the MCV EIA in detecting MCV antibodies suggesting antibody reactivity in this assay prima… Show more

“…Wild-type MCPyV may reside in skin and other tissues, with a high prevalence of infection in the general population (15,26,27). MCPyV-associated MCC is thought to arise in the rare event when MCPyV aberrantly integrates into the genome of the tumor progenitor cell and undergoes truncation or mutation of the LTAg gene that renders the virus replication-deficient but transformation-competent (2).…”

“…When expressed in mammalian or insect cells, VP1 proteins (or VP1 + VP2 proteins) self-assemble into 45-55 nm diameter virus-like particles (VLPs) (Fig. 2) that are used in serological assays (7,(12)(13)(14).…”

“…[5][6][7] Merkel cell polyomavirus has a high seroprevalence in the general population and asymptomatic infection begins in childhood. [8][9][10][11] As one of the steps in the proposed mechanism for Merkel cell carcinoma oncogenesis, polyomavirus must integrate into the human genome. 12 Viral integration sites occur throughout the human genome without apparent specificity.…”
