Human milk and mucosal lacto- and galacto-N-biose synthesis by transgalactosylation and their prebiotic potential in Lactobacillus species

Abstract: Lacto-N-biose (LNB) and galacto-N-biose (GNB) are major building blocks of free oligosaccharides and glycan moieties of glyco-complexes present in human milk and gastrointestinal mucosa. We have previously characterized the phospho-β-galactosidase GnbG from Lactobacillus casei BL23 that is involved in the metabolism of LNB and GNB. GnbG has been used here in transglycosylation reactions, and it showed the production of LNB and GNB with N-acetylglucosamine and N-acetylgalactosamine as acceptors, respectively. T… Show more

“…The disaccharides LNB, GNB, 3FN and 6FN were synthesized by transglycosylation reactions as previously described 20 , 21 , and they were tested independently in fermentation assays, using total microbiota isolated from stool samples of breastfed infants. The addition of any of those oligosaccharides decreased the culture pH (Table 1 ).…”

“…animalis and Bifidobacterium pseudolongum 22 . In addition, we have seen that LNB and GNB are also fermented by pure cultures of Lactobacillus species 20 . In this work, we have investigated, in batch cultures using total microbiota isolated from stool samples of breastfed infants, the effects of LNB, GNB, 3FN and 6FN on infant microbiota composition and subsequent short chain fatty acids production.…”

“…The gnb gene cluster is present in strains of the closely related species L. casei–paracasei–rhamnosus . These disaccharides are also metabolized by other species of intestinal lactobacilli ( L. gasseri and L. johnsonii ) although homologs of the gnb genes are not detected in their chromosomes, suggesting that as yet undiscovered pathways for the use of these mucosal-derived disaccharides are active in these bacteria ( Bidart et al, 2014 , 2017 ).…”
