Human Mn superoxide dismutase cDNA sequence

Abstract: Human Mn superoxide dismutase (MnSOD) encoded by chromosome 6 is a mitochondrial matrix enzyme positioned to scavenge oxygen radicals produced by the extensive oxidation-reduction and electron transport reactions undergoing in that organelle. cDNA clones containing the entire coding region for human MnSOD were isolated from a T-lymphocyte cDNA library in A gt1O. The cDNA contains a 666 bp coding region followed by a 3' untranslated region which lacks the AATAAA polyadenylation signal. The predicted amino acid … Show more

“…The cDNA for human mitochondrial MnSOD [15] was subcloned into the pcDNA3 expression vector in sense (MnSODsense) or antisense orientation (MnSODantisense) by standard molecular biology techniques [11]. RINm5F cells were stably transfected with the vector and positive clones were selected through resistance against G418 (250 μg/ml).…”

“…RINm5F tissue culture cells overexpressing Cat, Gpx, and Cu/ZnSOD were generated through stable transfection as described elsewhere (14). The full-length MnSOD cDNA fragment (18) was subcloned into the EcoRI site of the pcDNA3 plasmid, resulting in MnSOD sense and antisense expression constructs. The vector inserts were verified by sequence analysis.…”
