Human olfactory mesenchymal stromal cells co-expressing horizontal basal and ensheathing cell proteins in culture

Abstract: Introduction: The olfactory neuro-epithelium has an intrinsic capability of renewal during lifetime provided by the existence of globose and horizontal olfactory precursor cells. Additionally, mesenchymal stromal olfactory cells also support the homeostasis of the olfactory mucosa cell population. Under in vitro culture conditions with Dulbecco modified eagle/F12 medium supplemented with 10% fetal bovine serum, tissue biopsies from upper turbinate have generated an adherent population of cells expressing mainl… Show more

“…Assessment of the four markers by immunofluorescence microscopy (IMF, Figure D,E) was confirmatory of those obtained by flow cytometry. Hence, MSCs/MenSCs are naturally neurogenic. Since menstrual stromal cells (MenSCs) are well characterized and present minimal technical limitations, and minor ethical issues, this biological source cultured for 7 days in NeuroForsk medium has become one of the easiest methods to obtain a consistent, ready-to-use source of functional neurogenic cells to further investigate environmental and/or genetic aspects of PD. Therefore, we have used MenSC-derived dopaminergic-like neurons (DALNs) to inquire into the effect of CBD once exposed to the ambient pesticides PQ 2+ /MB, used as model neurotoxins in PD.…”

“…Harvesting techniques for OE in humans [ 28 , 29 , 30 , 43 , 44 ] and rats [ 45 ] are widely available; however they are scarce for mice [ 36 ]. This called for the development of our own harvesting surgical technique.…”

“…First, many studies about in vitro regrowth of olfactory axons use fetal bovine serum [ 28 ], containing a mixture of poorly characterized growth factors or Matrigel [ 29 ], a mixture of extracellular matrix proteins (ECM) [ 30 ]. Therefore, information on the effect of a single growth factor on axonal regrowth are scarce.…”

“…To induce neural differentiation, hO-MSCs were seeded in SFM supplemented with N2, RA and forskolin. In this regard, it was shown that SFM led to the differentiation of hO-MSCs into neurons and astroglial cells [ 96 ].…”
