Human Papillomavirus G-Quadruplexes

Abstract: Infection with human papillomaviruses (HPVs) is one of the most common sexually transmitted infections and can lead to development of head and neck, skin, and anogenital cancer, including cervical cancer, which represents one of the world's most significant health problems. In this study, we analyze G-rich regions in all known HPV genomes in order to evaluate their potential to fold into G-quadruplex structure. Interestingly, G-rich loci fulfilling the criteria for G-quadruplex formation were found in only 8 t… Show more

“…[137,139,141]. G-quadruplex structures were recognized, for example, in the Epstein-Barr virus-encoded nuclear antigen 1 (EBNA1) mRNA [142], as well as in all the known human papillomaviruses [143]. Moreover, G-quadruplex was also reported recently in the U3 region of the HIV-1 virus [144].…”

“…[12] Since s27 contains five G-rich tracts and has as such high potential to form Gquadruplex structures, itwas cut into two shorter oligonucleotides named s23 and s22, which comprise the first to the fourth and the second to the fifth G-tract, respectively ( Figure 1A). Thes equence-reversed oligonucleotides r27, r23, and r22 were also prepared.…”

“…Studies on G-quadruplexes in virus genomes are limited. A few studies show functional roles of selected RNA or DNA G-quadruplex motifs present in the virus genome in vitro [1317] or describe the genomic location of G-quadruplexes in virus genomes [18]. …”
