Human serum IgE‐mediated mast cell degranulation shows poor correlation to allergen‐specific IgE content

Abstract: Our studies demonstrate that mast cell activation mediated through IgE from allergic patients is a result of complex relationships that are not only dependent on allergen-specific IgE content but also relate to the capacity to efficiently sensitize and trigger the signalling responses that lead to degranulation.

“…To characterize saliva bioactivity, mast cells were incorporated into the minifeeders at the concentration of 10 6 /ml in Tyrode buffer and were exposed to mosquito bites (100 female mosquitoes) for 2 h. As controls, minifeeders containing the same mast cell preparations were kept out of reach of mosquitoes. Cells were harvested, and after centrifugation, supernatants were analyzed for their content in ␤-hexosaminidase as described previously (28). All results are expressed as a percentage of total ␤-hexosaminidase in the cells after correction for spontaneous release in unstimulated cultures (net release).…”

“…Dibbern et al (2003) showed that the absolute amount of peanut-specific IgE is an important determinant of the ability of a serum to effectively sensitize cells for allergen-dependent degranulation (r ¼ 0.95; p50.001), but that the amount of total IgE was a relatively poor predictor. Conversely, Marchand et al (2003) indicated no correlation (r ¼ 0.27) between the proportion of bound allergen-specific IgE and strength of the degranulation response after sensitization with serum IgE. However, a significant correlation (r ¼ 0.97) was found for purified monoclonal IgE.…”

“…Incubation with undiluted serum or 420% serum was found to cause cytotoxic effects (i.e. decrease viability) in the RBL cells (Ladics et al, 2008;Marchand et al, 2003). This toxicity appears to be due to naturally occurring xenoreactive antibodies (Palmera et al, 2005;Schuurman et al, 2003).…”

“…Upon activation by the high-affinity IgE receptors (FcRI), they release factors such as histamine, cytokines, chemokines that ultimately cause allergic responses (1,2). Gab2 is a downstream molecule of FcRI and plays an important role in FcRI-mediated signaling events in mast cells.…”
