Human T lymphocyte clones specific for malaria (Plasmodium falciparum) antigens.

Abstract: Peripheral blood mononuclear cells (PBM) from a patient who had lived in a malarial-endemic area were cultured in the presence of malarial antigens (a lysate of Plasmodium-infected erythrocytes). Responding cells were grown in IL-2-containing medium and then cloned, and subsequently subcloned, in the presence of phytohemagglutinin and allogeneic irradiated PBM. Ten clones were specific for malarial antigens. They proliferated in response to P. falciparum extract, but not to a lysate of uninfected erythrocytes.… Show more

“…Nevertheless, CD8 1 T cells specific for malaria antigens are activated during bloodstage malaria [10,11]. Taken together with our observation that the numbers of activated CD8 1 T cells increased after infection with PyL, antigens derived from malaria parasites must be presented on MHC class I molecules on APC including macrophages.…”

“…However, some studies have stressed the existence of CD8 1 T cells specific for blood-stage malaria parasites. First, CD8 1 T-cell clones that proliferate and produce IFN-g in response to blood-stage malarial antigens in an HLA-restricted manner are generated in patients who live in a malaria-endemic area [11]. Second, experimental infections of non-immune volunteers with an ultra-low dose of infected RBC induce immunity to subsequent challenge in the absence of detectable antibody responses.…”

“…We have earlier reported that both CD4' and CD8' P.5-specific TLC could be obtained from an acutely infected patient [9]. In this study TLC from another acutely infected and 4 clinically immune donors were analyzed.…”

“…PBMC from the patients were transformed with EBV as described [9]. Briefly, lo7 PBMC were resuspended in 10 ml RPMI-FCS containing 30% supernatant of the EBV-producing marmoset cell line B95.8 and 600 nglml cyclosporin A (Sandoz, Basel, Switzerland) and distributed in the wells of a flat-bottom 96-well microculture plate at 5 X 104/well.…”
