Human β-defensin-2 induction in Helicobacter pylori-infected gastric mucosal tissues: antimicrobial effect of overexpression

Abstract: The objective of this study was to understand more of the innate immune response to Helicobacter pylori by determining the expression of human â-defensin-2 (hBD-2) in various gastric mucosal tissues and MKN45 gastric cancer cells with or without H. pylori. Semi-quantitative TaqMan RT-PCR and immunohistochemistry were carried out. The antimicrobial effects of a transfected hBD-2 gene against H. pylori were also evaluated. The results showed that hBD-2 was expressed in inflamed gastric mucosal tissues with H. py… Show more

“…Statistical analysis was performed with a one-tailed t test (*, P Ͻ 0.05; **, P Ͻ 0.01). and trachea (3,52,53). The most prominent source of mBD-3 and mBD-14 assembly is likely stimulated nasal mucosa.…”

“…hBD-2 represents the first human defensin that is produced by epithelial cells following contact with bacteria, viruses, or cytokines, such as IL-1 and TNF-␣ (2,(17)(18)(19)(20)(21)(22). Although the nature of the regulation is not completely characterized, both the MAPK signal transduction pathway and the NF-B transcription factor have been suggested to be involved (23,24).…”

“…LL-37 is a cathelicidin with an anti-H. pylori effect [151] , and a recent study demonstrated that cathelicidin limited H. pylori colonization and related gastritis in mouse models [152] . Defensin peptides have also been indicated to impede H. pylori [153] . Human beta defensin 2 and 3 are differentially expressed in gastric mucosa during H. pylori infection [154] .…”
