Hydrogels of Dextran Containing Nonsteroidal Anti-Inflammatory Drugs as Pendant Agents

Abstract: The oral administration of nonsteroidal anti-inflammatory drugs (NSAIDs) is often associated with upper gastrointestinal tract side effects. To reduce these effects and improve the therapeutic efficacy, NSAIDs are often formulated as controlled release systems. We have prepared a new formulation consisting of dextran hydrogels containing NSAIDs as pendant agents, through ultraviolet irradiation of solutions of dextran functionalized with methacrylic groups in the presence of the drug derivatized in the same wa… Show more

“…The structure was characterized by 1 H NMR (Figure ). The peak assignments were in good agreement with that reported by Feeney et al The relative ratios of peak areas for protons a, b, c, d, and e were found to be 2.0:2.9:3.8: 2.8:3.7 (theoretical values are 2:3:4:3:4). 1 H, 13 C, and 2D NMR data are included in Figure S1 of the Supporting Information.…”

“…On the other hand, hydrolysis of the polymer backbone can also regulate drug release, perhaps stimulating the release of a relatively altered drug analogue. For instance, methacrylate-functionalized non-steroidal anti-inflammatory drugs have been conjugated to methacrylic functionalized dextrans by means of UV radiation; a chemically modified drug analogue is released as the dextran hydrogel degrades [151,152].…”

“…Therefore, some advanced delivery systems have been investigated to address the formulation limitations of currently available anti‐inflammatory drugs . The examined new pharmaceutical strategies include hydrogels, microparticles, nanofibers, lipid/polymeric vesicles, polymeric or lipid nanoparticles, micelles, nanocapsules, micro‐ or nano‐emulsions, and polymer‐drug conjugates, among others. In particular, nanotherapies have attracted considerable attention for the treatment of different acute and chronic inflammatory diseases, such as acute liver/lung injury, cardiovascular disease, neurodegenerative disorders, and inflammatory bowel disease .…”
