Hepato-protective effects of Montelukast against methotrexate-induced hepatotoxicity in rat model

Aim: Assessment of the possible Hepato-protective effects of Montelukast against hepatotoxicity induced by MTX in rat model. Materials and methods: A 24 Albino Wister rats were included in this study, they were randomly divided into 4 groups each of 6. Group 1: control group (administered 0.9% N/S 1ml/kg/d ip) for 5 days. Group 2: MTX group (received MTX in a dose of 20mg/kg/day ip) as a single dose at day 1 and observed till day 5 of the experiment. Group 3: Montelukast group (received MTX in a dose of 20mg/kg/d ip as a single dose then Montelukast 10 mg /kg/ d ip for 5days). Group 4: vehicle group received 1ml/kg ip for each rat (10%DMSO + 40%PEG300 + 5% Tween400 + 45% Normalsalin) vehicle solution for 5 days. At day 5 all animals were sacrificed, serum was aspirated for the assessment of ALT, AST, ALP, TSB levels by colorimetric assay. Liver was removed and divided into 2 parts for histopathological examination and for assessment of tissue TNF-alpha, caspase-3, MDA, total antioxidant capacity, and TLR4 levels by ELISA method. Results: Mean serum levels of ALT, AST, ALP and TSB as well as tissue MDA, TNFα, TlLR4, Caspse3 were significantly increased in MTX group compared with the control and vehicle groups together with significant reduction in total antioxidant capacity and obvious histological damage. Whereas meanwhile treatment of rats with Montelukast together with MTX resulted in significant reduction in mean serum levels of ALT, AST, ALP and TSB as well as tissue MDA, TNFα,TlLR4, Caspse3 together with significant increment in total antioxidant capacity and near normal restoration of hepatic tissue architecture. Conclusion: Montelukast has Hepatoprotective effect against MTX induced hepatotoxicity by its antioxidant, anti-inflammatory, and anti-apoptotic properties.

Seed Endophytic Bacteria of Pearl Millet (Pennisetum glaucum L.) Promote Seedling Development and Defend Against a Fungal Phytopathogen

Seed endophytic bacteria (SEB) are primary symbionts that play crucial roles in plant growth and development. The present study reports the isolation of seven culturable SEB including Kosakonia cowanii (KAS1), Bacillus subtilis (KAS2), Bacillus tequilensis (KAS3), Pantoea stewartii (KAS4), Paenibacillus dendritiformis (KAS5), Pseudomonas aeruginosa (KAS6), and Bacillus velezensis (KAS7) in pearl millet seeds. All the isolates were characterized for their plant growth promoting activities. Most of the SEB also inhibited the growth of tested fungal phytopathogens in dual plate culture. Removal of these SEB from seeds compromised the growth and development of seedlings, however, re-inoculation with the SEB (Kosakonia cowanii, Pantoea stewartii, and Pseudomonas aeruginosa) restored the growth and development of seedlings significantly. Fluorescence microscopy showed inter and intracellular colonization of SEB in root parenchyma and root hair cells. Lipopeptides were extracted from all three Bacillus spp. which showed strong antifungal activity against tested fungal pathogens. Antifungal lipopeptide genes were also screened in Bacillus spp. After lipopeptide treatment, live-dead staining with fluorescence microscopy along with bright-field and scanning electron microscopy (SEM) revealed structural deformation and cell death in Fusarium mycelia and spores. Furthermore, the development of pores in the membrane and leakages of protoplasmic substances from cells and ultimately death of hyphae and spores were also confirmed. In microcosm assays, treatment of seeds with Bacillus subtilis or application of its lipopeptide alone significantly protected seedlings from Fusarium sp. infection.

Diversity and Exploration of Endophytic Bacilli for the Management of Head Scab (Fusarium graminearum) of Wheat

Fusarium graminearum causing head scab (HS) or head blight (HB) disease in wheat is one of the nasty fungi reported to cause significant grain quality and yield loss. Biological control using endophytic bacteria has emerged as a prospective option for containing fungal diseases in an environmentally benevolent, durable, and sustainable manner. In this regard, 112 endophytic bacilli were isolated from the anthesis stage (Zadok’s growth stage 65) from five different wheat genotypes with an aim to identify prospective antagonistic strains against F. graminearum. The molecular identity of the strains was confirmed by matching 16S rRNA sequences of bacterial strains with the gene sequences of type strains available in the National Center for Biotechnology Information database and reported 38 different species of Bacillus in all the five wheat cultivars. Further, it has been observed that only fourteen strains (B. clarus NOK09, B. mojavensis NOK16, B. subtilis NOK33, B. rugosus NOK47, B. mojavensisNOK52, B. clarus NOK59, B. coahuilensis NOK72, B. cabrialesii NOK78, B. cabrialesii NOK82, B. rugosus NOK85, B. amyloliquefaciens NOK89, B. australimaris NOK95, B. pumilus NOK103, and B. amyloliquefaciens NOK109) displayed in-vitro antagonistic effect against Fusarium graminearum fungus. Furthermore, the three endophytic Bacillus strains showing the strongest antagonistic effect (> 70% of growth inhibition of fungal mycelium) under in-vitro antagonistic assay were selected for field experiments. In a two-year consecutive field study, a combination of three strains (B. clarus NOK09 + B. subtilis NOK33 + B. amyloliquefaciens NOK109) displayed a remarkable reduction in HS disease index by 81.47% and 77.85%, respectively. Polymerase chain reaction assay detected three genes (ituD, bmyC, and srfA) involved in antibiotic biosynthesis pathways. Additional attributes such as potassium solubilization, siderophore release, and hydrolytic enzyme (protease, lipase, amylase, chitinase, and pectinase) synthesis have been observed in these strains. Overall, the present study was successful in profiling endophytic bacilli and selecting the combination of effective antagonistic endophytic Bacillus strains that could be the best alternative for the sustainable and ecological sound management of HS disease in wheat under field conditions.
