Hydrostatic countercurrent chromatography and ultra high pressure LC: Two fast complementary separation methods for the preparative isolation and the analysis of the fragrant massoia lactones

Abstract: Using a one-step preparative hydrostatic countercurrent chromatography method, the fragrant massoia lactones were purified from the crude massoia bark oil, in less than 3 h. The fractionation was performed with the biphasic solvent system c-hexane-methanol-water (10:9:1, v/v/v), leading to target compounds with purity over 96%, as determined by GC-MS and ultra high pressure LC-MS analyses. Together with C-10, C-12 and C-14 massoia lactones, two other aromatic compounds used in perfumes, benzyl benzoate and ben… Show more

“…Additionally, massoia lactone was the dominant component in JSBC volatiles (accounting for 77.46% of the total peak areas) and thus could be regarded as a marker for quality control of this product ( Table 2 ). Massoia lactone is a fragrant agent that may be sensed as coconut, cream or butter, and is therefore used in food industry as a flavor additive [ 26 ]. Recent studies suggest that massoia lactone exhibits potential antifungal, antivirus, anticancer, or anti-inflammatory activities, which might contribute to the pharmacological effects of JSBC [ 25 , 27 ].…”

“…The massoia tree is native to the rainforests of Indonesia and Papua New Guinea 22 . Its wood and bark are used in perfumes, food, and traditional medicine [22][23][24] . Biological activities of massoia EO, such as immunomodulatory effects, antifungal and phytotoxic activity have been reported 22,24,25 .…”

“…albopictus may be attributable to the methodological differences, as bin Janten et al 37 used fourth instar larvae for larvicidal tests and ethanol as a solvent for EOs, unlike the method of this study. C10 and C12 massoia lactones are reportedly the main constituents of massoia EO 23,33 , and they possess an α-β-unsaturated δ-lactone moiety. Massoia lactone groups have been reported to show biological activities, including antimicrobial, cytotoxic, anti-inflammatory and phytotoxic activities 22,24,43,44 .…”
