Hypo-Osmotic Swelling Test (HOST) for Feline Spermatozoa: The Simplified Procedure and the Aspect of Sperm Morphology

Abstract: Hypo-osmotic swelling test (HOST) is used to assess the functional integrity of sperm plasma membranes in many species. The primary aim of this study was to test a simplified HOST procedure for the evaluation of feline semen. The second objective was to check if sperm abnormalities can influence the results of this test. Urethral semen was collected from 19 male, domestic cats. In Exp. 1, HOST was performed in different media (50 mOsm/kg fructose or distilled water), temperature (37 °C or room temperature) and… Show more

“…The toxic effects of ROS were lipid peroxidation, protein ionization and inactivation, and DNA damage (Juan et al, 2021). After the addition of the hypo-osmotic solution, the damaged spermatozoa plasma membrane is seen straight tail shape (Prochowska et al, 2022).…”

“…The thawed sperm membrane integrity was evaluated by a hypo-osmotic swelling test (HOST) [ 22 ]. A 20 µL aliquot of thawed semen was diluted in 200 µL of hypo-osmotic solution (9 mg/mL fructose and 4.9 mg/mL sodium citrate).…”

“…Hypo-osmotic swelling test (HOST) was used to measure the sperm plasma membrane integrity [65]. Briefly, 200 µL of the semen was mixed with 1 mL of HOST solution and incubated at 37 • C for 30 min.…”
