Hypoglycemia Induces Tau Hyperphosphorylation

Abstract: Cerebral hypoglycemia/hypometabolism is associated with Alzheimer's disease (AD) and is routinely used to assist clinical diagnosis of AD by brain imaging. However, whether cerebral hypoglycemia/hypometabolism contributes to the development of AD or is a response of reduced neuronal activity remains unclear. To investigate the causal relationship, we cultured the differentiated N2a neuroblastoma cells in glucose/pyruvate-deficient media (GDM). Shortly after the N2a cells cultured in the GDM, the mitochondria m… Show more

“…A decrease in glucose metabolism in the hypoxic and hypoperfused brain may trigger the process of neuronal degeneration in AD. Hypoglycemia increases neuronal tau hyperphosphorylation (Lee et al, ). Intracerebroventricular injection of streptozotocin (STZ) in Wistar rats selectively affected glucose transporter type 2‐bearing cells (astrocytes) and interrupted glucose supply to neurons.…”

“…LX2343 ameliorated learning and memory impairments in ICV-STZ rats ICV injection of rats with a sub-diabetogenic dose of STV has been shown to be closely linked to the sporadic dementia of Alzheimer's disease [34] , which is characterized by cognitive impairment [35] , OS [35] , impaired glucose metabolism [36] , and hyperphosphorylated tau [37] . Given the pathological features of ICV-STZ rats and the efficiency of LX2343 in both protecting neuronal cells and attenuating tau pathology, we performed the MWM assay to examine the potential of LX2343 in the amelioration of memory impairment in ICV-STZ rats.…”

“…There are very few arguments about the role of abnormal tau hyperphosphorylation in AD, related tauopathies and under experimental conditions [108, 109, 114118]. A subject to be explored in air pollution animal models ought to be the characterization of the HP τ and if indeed represents a compensatory neuronal response against oxidative stress.…”
