Hypotaurine Is a Substrate of GABA Transporter Family Members GAT2/Slc6a13 and TAUT/Slc6a6

Abstract: Hypotaurine is a precursor of taurine and a physiological antioxidant that circulates in adult and fetal plasma. The purpose of the present study was to clarify whether hypotaurine is a substrate of Slc6a/gammaaminobutyric acid (GABA) transporter family members. Radiolabeled hypotaurine was synthesized from radiolabeled cysteamine and 2-aminoethanethiol dioxygenase. The uptakes of [ 3 H]GABA, [ 3 H]taurine, and [ 14 C]hypotaurine by HEK293 cells expressing mouse GAT1/Slc6a1, TAUT/Slc6a6, GAT3/Slc6a11, BGT1/ Sl… Show more

“…6). These two compounds are related to the gene SLC6A13 as either substrates (hypotaurine) or derivative of a substrate (N_methyl_GABA) [11]. Mouse knockouts of SLC6A13 are known to have 50% lower taurine levels in the liver compared to wildtype individuals [12].…”

“…As a taurine precursor, hypotaurine is produced by cysteine. At high-LIN treatment groups, genes cysteine dioxygenase (cdo) and cysteamine dioxygenase (ado) involved in two ways of producing cysteine were both downregulated [55]. Besides, hypotaurine is an endogenous antioxidant, the decrease of these two genes ado and cdo1 suggested a reduction of intracellular hypotaurine level, which is proof of cellular responses to oxidative stress [56].…”
