Hypusination of eukaryotic initiation factor 5A via cAMP-PKA-ERK1/2 pathway is required for ligand-induced downregulation of LH receptor mRNA expression in the ovary

Abstract: Luteinizing hormone receptor (LHR) mRNA expression in the ovary is regulated post-transcriptionally by an LH receptor mRNA binding protein (LRBP). Eukaryotic initiation factor 5A (EIF5A), identified as an LRBP-interacting protein plays a crucial role in LHR mRNA expression. In this study, we have demonstrated that during hCG-induced LHR downregulation, a significant upregulation of eIF5A mRNA expression and hypusination of eIF5A protein occurs in a time dependent manner. Pretreatment with H89, a specific inhib… Show more

“…For example, a total of 29 mRNAs were involved in cAMP signaling. cAMP signaling has previously been implicated that it was expressed in mature follicles to maintain oocyte meiosis inhibition in ovarian (Gulappa et al, 2015;Lyga et al, 2016). In particular, the downstream genes of PTCH1, GLI1, HHIP, and AMH were found to be upregulated in small follicles, suggesting that these genes are implicated in Hedgehog and AMH transduction in small follicles.…”

“…Among them, RPL10A, RPL23A, RPL24, RPL34, RPL37, RPLP0, RPS3A , RPS6 and RPS7 are all common protein family ribosomal protein, they play an important role in the development of ovarian and breast cancers related to precocious puberty [38,39]. And EIF5A, HNRNPDL, HSPA8, PKN1, PRKAR2B, SLC40A1, SLC25A5, SNRPD2 and YWHAH, play key roles in oocyte meiosis, steroid biosynthesis and conduction of reproduction process [40][41][42].…”

“…It has been revealed that eIF5A plays an important role in regulating cellular autophagy. The mRNA level of eIF5A was significantly upregulated after hCG-induced downregulation of LHR, whereas eIF5A level decreased over time, suggesting that eIF5A is involved in the post-transcriptional regulation of ovarian LHR mRNA-binding protein [55]. Therefore, we hypothesized that spermidine mediates hyp-eIF5A modification and regulates ovarian autophagy to inhibit ovarian aging, but in-depth studies are needed to unravel the specific mechanisms.…”
