<i>ADRB2</i> polymorphisms in allergic asthma in Han Chinese children

Regulation of the immune response in asthma is complex. MicroRNA-126 (miR-126) expression has been implicated in this response, so we sought to determine the clinical significance of miR-126 measured in the peripheral blood. A total of 80 children with acute asthma were selected to participate in the study and were compared to 80 healthy children. The relative circulating miR-126 levels, interleukin (IL)-4 levels, and the Th17 cell percentage in the peripheral blood of children in the case group were significantly higher than those in the control group, while the interferon (IFN)-γ levels and the CD4CD25Treg cell percentage were significantly lower than those in the control group. Along with the aggravation of the disease, the relative levels of miR-126 and IL-4 and the percentage of Th17 cells increased gradually, while the IFN-γ levels and the CD4CD25Treg cell percentage decreased. The relative level of miR-126 in the peripheral blood of children with asthma was positively correlated with IL-4 and the Th17 cell percentage and was negatively correlated with IFN-γ levels, CD4CD25Treg cell percentage and lung function indicators. The relative level of miR-126 was correlated with the Th17 cell percentage in the peripheral blood, forced vital capacity (FVC), and forced expiratory flow (FEF) of the children with asthma. The relative levels of miR-126 and IL-4 and the Th17 cell percentage were positively correlated with the severity of the asthma, while IFN-γ levels and the CD4CD25Treg cell percentage were negatively correlated with the severity of the asthma. CD4CD25Treg cell percentage and relative miR-126 levels were of the most predictive value in the diagnosis of asthma. Our findings show that the overexpression of miR-126 in acute asthma is correlated with signs of immune imbalance and is predictive of the severity of the disease, suggesting that it could be used as a potential serological marker for asthma diagnosis and evaluation.

were searched using terms: ("asthma" or "asthmatic") and ("beta-2 adrenergic receptor" or "ADRB2" or "b2-AR") and "polymorphism" and ("Chinese" or "China"). The search had no restriction in language or publication status.Inclusion standards: (1) studies examining the relationship between ADRB2 Arg16Gly polymorphism and asthma, (2) study design with the case-control, (3) sufficient data of genotype frequency, (3) Chinese individuals. Exclusion standards: (1) overlapped literatures, (2) unextractable data, (3) not a case-control design, (4) abstract or reviews. The potentially relevant studies and extracted data from the identified publications, including first author's name, publication year, geographic areas, age group, sample size, and available genotype information from ADRB2 Arg16Gly polymorphism were screened by the investigators. Statistical analysisData were analyzed by STATA version 12 (StataCorp LP, College Station, TX, USA). Odds ratios (ORs) with 95% confidence intervals (CIs) were calculated and analyzed by Z-test. The model of G versus A, GG versus AA, GG versus (AA+AG) and (GG+AG) versus AA were examined with the asthma risk, respectively. Heterogeneity and Hardy-Weinberg equilibrium (HWE) were evaluated by I-squared based on Q and the df. When P heterogeneity < 0.1 or I 2 > 50%, a random-effects meta-analysis model was performed to estimate the pooled ORs; if not, a fixed-effects model was used. Pooled ORs were collected for the sensitivity assay. A p-value < 0.05 showed the significant change.

Поліморфізм гена ADRB2 як чинник спадкової схильності до розвитку бронхіальної астми та відповіді на терапію сальбутамолом 1І нститут молекулярної біології і генетики НАН України, м. Київ Мета -дослідити асоціацію поліморфізму C79G гена АDRB2 із ризиком розвитку бронхіальної астми (БА) у дітей, що мешкали в різних умовах забруднення навколишнього середовища, а також змоделювати 3D структури ізоформ білка β2 адренорецептора, що кодуються поліморфними варі антами гена АDRB2 (A46G, C79G, та C491T) in silico, для передбачення структурно функціональних властивостей, які можуть впливати на взаємодію із сальбутамолом. Пацієнти та методи. Обстежено 114 дітей віком від 3 до 18 років із БА середньої тяжкості, контрольованого перебігу, які методом рандомізації були розподілені на дві групи: групу І (діти з умовно чистого регіону Києва та Київської області) та групу ІІ (діти з екологічно забрудненого регіону). До групи контролю увійшли 86 неспоріднених здорових дорослих із різних регіонів України. Поліморфний варіант гена АDRB2 (C79G) досліджено методом алельспецифічної полімеразної ланцюгової реакції. Комп'ютерне моделювання за гомологією 3D структури білка β2 адренорецептора проведено з використанням вебсерверів SWISS MODEL та I TASSER, молекулярний докінг здій снено за допомогою програми Auto Dock Vina. Результати. Встановлено, що частота носіїв поліморфної алелі 79G гена ADRB2 є статистично вірогідно вищою (p<0,05) у групі обстеження ІI (69,4%) порівняно з частотою (55,8%) у контрольній групі. За результатами аналізу змодельованої просторової структури білка ADRB2, заміни p.16Arg>Gly та p.27Gln>Glu знаходяться в N кінцевій послідовності і можуть впливати на взаємодію з білками партнерами, у свою чергу, амінокислотна заміна p.164Thr>Ile локалізована поблизу сайту зв'язування з лігандами і може знижувати афінність сальбутамолу до відповідного мутантного рецептора. Висновки. Поліморфний варіант 79G гена ADRB2 можна розглядати як фактор спадкової схильності розвитку БА в умовах антропогенного навантаження навколишнього середовища. Мононуклеотидну заміну 491C>T гена ADRB2 можна розглядати як фармакогенетичний маркер поганої відповіді пацієнта на лікування сальбутамолом.
