Arabidopsis thaliana inositol 1,3,4‐trisphosphate 5/6‐kinase 4 (AtITPK4) is an outlier to a family of ATP‐grasp fold proteins from Arabidopsis

Abstract: The Arabidopsis genome encodes a family of inositol 1,3,4-trisphosphate 5/6-kinases which form a subgroup of a larger group of ATP-grasp fold proteins. An analysis of the inositol 1,3,4-trisphosphate 5/6-kinase family might, ultimately, be best rewarded by detailed comparison of related enzymes in a single genome. The enzyme encoded by At2G43980, AtITPK4; is an outlier to its family. At2G43980 is expressed in male and female organs of young and mature flowers. AtITPK4 differs from other family members in that … Show more

“…soybean Itpks The canonical catalytic activity of all Itpk proteins is the phosphorylation of Ins(1,3,4)P 3 to both Ins(1,3,4,5)P 4 and Ins(1,3,4,6)P 4 [12,14,20,21]. We found that all four soybean Itpks phosphorylated Ins(1,3,4)P 3 ( Fig.…”

“…Little work has been done to identify enzyme(s) that might catalyze these reactions, with the exception of a recent report showing that a racemic mixture of Ins(3,4,6)P 3 /Ins(1,4,6)P 3 was phosphorylated by the recombinant Itpk from Arabidopsis [14]. The use of a substrate mixture prevents us from knowing which of the two were phosphorylated.…”

“…Ins(3,4,5,6)P 4 was identified in early studies of IP 6 synthesis in duckweed (50). This isomer is a substrate of plant ATP-grasp fold inositol tris/tetrakisphosphate kinases (20,(51)(52)(53), and enzymes of this class contribute to IP 6 synthesis in maize (20). We have shown using a coupled assay that Ins(3,4,5,6)P 4 and Ins(1,4,5,6)P 4 are equally effective in vitro substrates for IP 5 2-K, and we have now determined the role of Arg 130 and Arg 45 in the specification of substrate preference between enantiomers.…”

“…ucts, IPK1 has a dominant contribution to inositol hexakisphosphate accumulation (25). We undertook expression analysis of the effect of mutation of AtMRP5 on transcript levels of AtIPK1 and of four members of the inositol tris/tetrakisphosphate kinase family whose contribution to inositol hexakisphosphate synthesis, while undefined in Arabidopsis (34), is anticipated from studies of maize (30). The data presented in the supplementary information (supplemental Fig.…”
