P53 Codon 72 Polymorphism and Risk for Squamous Cell Carcinoma of the Penis: A Caucasian Case-Control Study

Abstract: Squamous cell carcinoma of the penis is a rare but often aggressive disease. A large proportion of penile cancers are associated with HPV infection, mainly with HPV high-risk subtypes 16 and 18. From other HPV-related malignancies a link between a functional SNP in the p53 gene (rs1042522, p.Arg72Pro) and a higher disease risk in the presence of HPV is documented. The p53 p.Arg72 variant was described as a risk factor for developing a malignancy in combination with the presence of HPV as the p.72Arg variant is… Show more

“…Mutant TP53 has an influence on the proliferation, migration, survival, and invasion of tumors and the drug resistance of chemotherapy drugs ( 34 ). When TP53 is mutated, the original tumor suppressor function is affected, which further increases the apoptosis process of gastric mucosa cells and impairs the damage repair function of the cells, thus inducing the transition of normal cells to cancer cells and increasing the incidence of GC ( 35 ). ATM is a cell housekeeping gene encoding ATM protein kinase, which plays a key role in the early signal transduction of cell cycle checkpoints ( 36 , 37 ).…”

“…All tumors of the cohort were classified and staged according to the World Health Organization classification of penile tumors and the current American Joint Committee on Cancer/TNM-classification system (22,23). The cases analyzed are part of a larger cohort described previously (24). The clinicopathological characteristics of the subset of cases used in the present study are presented in Tables I and II.…”

“…Microdissection of tissue and DNA isolation. Microdissection and isolation of genomic DNA from FFPE tissue were performed as described previously (24,25). As a prerequisite, serial tissue sections (5 µm thickness) were dewaxed, rehydrated and stained with 0.1% methylene blue (15 sec at room temperature).…”
