Scutellaria barbata D. Don (SBD) protects oxygen glucose deprivation/reperfusion-induced injuries of PC12 cells by up-regulating Nrf2

Abstract: This study aimed to investigate the potential effect of Scutellaria barbata D. Don (SBD) on oxygen glucose deprivation/reperfusion (OGD/R)-injured PC12 cells. PC12 cells were pretreated with various concentrations of 0.1-0.8 mg/ml SBD for indicated times (12-48 h) and then subjected to OGD/R injury. Cell viability, apoptosis and proliferation were detected using MTT assay, flow cytometry, Ki67 staining and western blot. Oxidative damage was assessed by detecting MDA content, SOD activity and GSH levels. The mi… Show more

“…Antioxidant, mitochondrial health and anti-apoptosis are always interrelated in overcoming oxidative stress-induced toxicity, hence becoming the main targets of neuroprotective therapy [72,73,74,75,76]. Similarly, we found in this study that the increase in antioxidant activities were accompanied by improvement of MMP and reduction of mitochondrial toxicity.…”

“…Moreover, our in vitro data also indicated that EA treatment suppressed OGD/R-induced ROS generation in primary cortical neurons. MDA, a final breakdown product of lipid peroxidation, is used to measure the extent of oxidative damage to cell membranes ( Wang et al, 2019 ). Our study showed that EA reduced the level of MDA in the ipsilateral hemisphere of neonatal mice following HIBD.…”

“…In addition, Lv's data indicated that CXCL8 gene silencing accelerated the activation of neuroglial cells while suppressing neuroinflammation via the PI3K/Akt/NF-κB pathway in the mice of CIS (50). Besides, LY294002, as a PI3K inhibitor that is widely used for the study of the characteristics of PI3K cell signaling pathways (51)(52)(53), inhibited the expressions of PI3K/Akt/NF-κB pathway and blocked the function of SEH treatment on PC12 cells injured by OGD/R. For instance, published study indicated that ginkgolides protected against ischemia-reperfusion damage in vivo and in vitro through the activation of Nrf2 and CREB via PI3K/Akt signaling, which could be reversed by cotreatment with LY294002 (54).…”
