Ibis T5000: a universal biosensor approach for microbiology

Abstract: We describe a new technology, the Ibis T5000, for the identification of pathogens in clinical and environmental samples. The Ibis T5000 couples nucleic acid amplification to high-performance electrospray ionization mass spectrometry and base-composition analysis. The system enables the identification and quantification of a broad set of pathogens, including all known bacteria, all major groups of pathogenic fungi and the major families of viruses that cause disease in humans and animals, along with the detecti… Show more

“…All groups of bacteria, including intracellular organisms such as Mycoplasma, Chlamydia, and Rickettsia and hard to culture or nonculturable organisms, could be detected by this assay. Analysis was performed using the software version 2.6.052 (Ibis Biosciences; Ecker et al, 2008). The PLEX-ID BAC Detection assay has a DNA calibrant in each reaction, which allows for semiquantitative analysis (Hofstadler et al, 2005).…”

“…This technology consists of DNA amplification by PCR of specific regions of ribosomal and conserved house-keeping genes, as well as some antibiotic-resistant genes, which are electrosprayed into a time-of-flight MS for molecular weight measurement. The mass of each amplicon is translated into base composition for organism and antibiotic-resistant gene identification (see Ecker et al, 2008, andWolk et al, 2012, for reviews of the methodology, flow scheme, and laboratory application). This technology has also found applications in human clinical diagnostics (Arciola et al, 2011;Wolk et al, 2012).…”

“…Cette approche, qui s'applique directement au prélèvement, permet de s'affranchir de l'étape de culture. Elle est rapide (résultats en quelques heures), semi-quantitative et détecte un large panel de micro-organismes (bactéries, champignons et virus) [4,5]. Il faut cependant en évaluer la sensibilité et la spécificité.…”

“…The system is based on base ratios (not base sequences) and can provide test results in six to seven hours (Ecker et al, 2008). The method was recently identified as the molecular system most likely to fulfill the requirements of routine diagnosis in orthopedic surgery (Costerton et al, 2011).…”
