Identification and functional characterization of Trypanosoma brucei peroxin 16

Abstract: Protozoan parasites of the family Trypanosomatidae infect humans as well as livestock causing devastating diseases like sleeping sickness, Chagas disease, and Leishmaniasis. These parasites compartmentalize glycolytic enzymes within unique organelles, the glycosomes. Glycosomes represent a subclass of peroxisomes and they are essential for the parasite survival. Hence, disruption of glycosome biogenesis is an attractive drug target for these Neglected Tropical Diseases (NTDs). Peroxin 16 (PEX16) plays an essen… Show more

“…Additionally, a Pex16 homolog was recently identified in T. brucei and silencing of this glycosome protein results in glycosome abnormalities with accumulation of structures in the anterior part of the cells where ER exit sites are located (Kalel et al. ).…”

“…The mechanisms of glycosome biogenesis are not fully understood, although comparison of components required for peroxisome biosynthesis has been useful in resolving potential contributing proteins (Galland and Michels ; Kalel et al. ). For example, homologs of 10 of the more than 58 described eukaryotic peroxin (Pex) proteins have been identified in T. brucei .…”

“…A considerable number of T. brucei homologs of mammalian and yeast peroxins involved in PMP insertion and matrix protein import have been identified and characterized, and shown to be involved in glycosome biogenesis (Kalel et al, 2015(Kalel et al, , 2019Banerjee et al, 2019;and reviewed in Galland and Michels, 2010;Gualdrón-López et al, 2013). Orthologous genes have been detected in the genome of T. cruzi and several of the proteins in its glycosomal proteome (Acosta et al, 2019).…”

“…The biogenesis of peroxisomes and glycosomes can involve new organelles that arise from pre-existing peroxisomal/glycosomal structures or de novo synthesis from the endoplasmic reticulum. However, in eukaryotes subject to environmental change, peroxisome composition can be modulated to allow metabolic adaptation, and this can be achieved by de novo loading from the ER (27) as well as the growth and division of existing glycosomes. The dynamics of environmental adaptation in procyclic form parasites have been analysed both during differentiation and upon exposure of procyclic forms to low and high glucose concentrations.…”
