With Next Generation Sequencing technologies, high-throughput RNA sequencing (RNAseq) was conducted to examine gene expression in neonates of Diabrotica virgifera virgifera (LeConte) (Western Corn Rootworm, WCR) challenged with individual proteins of the binary Bacillus thuringiensis insecticidal proteins, Cry34Ab1 and Cry35Ab1, and the combination of Cry34/Cry35Ab1, which together are active against rootworm larvae. Integrated results of three different statistical comparisons identified 114 and 1300 differentially expressed transcripts (DETs) in the Cry34Ab1 and Cry34/35Ab1 treatment, respectively, as compared to the control. No DETs were identified in the Cry35Ab1 treatment. Putative Bt binding receptors previously identified in other insect species were not identified in DETs in this study. The majority of DETs (75% with Cry34Ab1 and 68.3% with Cry34/35Ab1 treatments) had no significant hits in the NCBI nr database. In addition, 92 DETs were shared between Cry34Ab1 and Cry34/35Ab1 treatments. Further analysis revealed that the most abundant DETs in both Cry34Ab1 and Cry34/35Ab1 treatments were associated with binding and catalytic activity. Results from this study confirmed the nature of these binary toxins against WCR larvae and provide a fundamental profile of expression pattern of genes in response to challenge of the Cry34/35Ab1 toxin, which may provide insight into potential resistance mechanisms.

The susceptibility of adults and larvae of the Japanese beetle to isolates of the fungi Beauveria bassiana, B. brongniartii, Metarhizium anisopliae, Lecanicillium longisporum and L. muscarium (Ascomycota: Hypocreales) was evaluated in laboratory bioassays. The presence of intra- and interspecific variations regarding the pathogenicity of the isolates was investigated. Results show that most of the fungal isolates caused mortality in adults and larvae. Isolates M. anisopliae INRS 705 and B. bassiana INRS 236 induced 70.3% and 65.2% of mortality in P. japonica adults, respectively, and both caused the death of about 37% of larvae. Of the 17 tested isolates, 13 caused larval mortality. However, no significant difference was found between the susceptibility of larvae to isolates from different species and those from a same species. Lecanicillium species are undoubtedly not pathogenic to adults and seem to have few effects on larvae. Based on the results obtained with the selected isolates, intra- and interspecific differences relative to the pathogenicity of the isolates appeared to be present. Overall, this study expanded the knowledge about P. japonica susceptibility towards entomopathogenic Hypocreales fungi. The implications of this study regarding the development of a biological control agent are discussed.La vulnérabilité des adultes et des larves du scarabée japonais aux isolats des champignons Beauveria bassiana, B. brongniartii, Metarhizium anisopliae, Lecanicillium longisporum et L. muscarium (Ascomycota : Hypocreales) a été évaluée lors de tests biologiques en laboratoire. La présence de variations intra- et interspécifiques concernant le pouvoir pathogène des isolats a été étudiée. Les résultats montrent que la plupart des isolats fongiques provoquent de la mortalité chez les adultes et les larves. Les isolats M. anisopliae INRS 705 et B. bassiana INRS 236 ont mené à des taux de mortalité de 70,3 % et 65,2 % chez les adultes de P. japonica, respectivement, et les deux isolats ont provoqué la mort d’environ 37 % des larves. Cependant, aucune différence importante n’a été notée entre la vulnérabilité des larves aux isolats de différentes espèces et à ceux de la même espèce. Les espèces Lecanicillium n’ont clairement pas de pouvoir pathogène chez les adultes et semblent n’avoir que peu d’effet chez les larves. En se fondant sur les résultats obtenus avec les isolats choisis, des différences intra- et interspécifiques concernant le pouvoir pathogène des isolats semblent être présentes. Directed evolution methods mimic in vitro Darwinian evolution, inducing random mutations and selective pressure in genes to obtain proteins with enhanced characteristics. These techniques are developed using trial-and-error testing at an experimental level with a high degree of uncertainty. Therefore, in silico modeling of directed evolution is required to support experimental assays. Several in silico approaches have reproduced directed evolution, using statistical, thermodynamic, and kinetic models in an attempt to recreate experimental conditions. Likewise, optimization techniques using heuristic models have been used to understand and find the best scenarios of directed evolution. Our study uses an in silico model named HeurIstics DirecteD EvolutioN, which is based on a genetic algorithm designed to generate chimeric libraries from 2 parental genes, cry11Aa and cry11Ba, of Bacillus thuringiensis. These genes encode crystal-shaped δ-endotoxins with 3 conserved domains. Cry11 toxins are of biotechnological interest because they have shown to be effective as biopesticides for disease-spreading vectors. With our heuristic model, we considered experimental parameters such as DNA fragmentation length, number of generations or simulation cycles, and mutation rate, to get characteristics of Cry11 chimeric libraries such as percentage of population identity, truncation of variants obtained from the presence of internal stop codons, percentage of thermodynamic diversity, and stability of variants. Our study allowed us to focus on experimental conditions that may be useful for the design of in vitro and in silico experiments of directed evolution with Cry toxins of 3 conserved domains. Furthermore, we obtained in silico libraries of Cry11 variants, in which structural characteristics of wild Cry families were observed in a review of a sample of in silico sequences. We consider that future studies could use our in silico libraries and heuristic computational models, as the one suggested here, to support in vitro experiments of directed evolution.
