Identification of a Prognostic Model Based on Fatty Acid Metabolism-Related Genes of Head and Neck Squamous Cell Carcinoma

Abstract: The fatty acid metabolism (FAM) is known to impact tumorigenesis, tumor progression and treatment resistance via enhancing lipid synthesis, storage and catabolism. However, the role of FAM in head and neck squamous cell carcinoma (HNSCC) has remained elusive. In the present study, we obtained a total of 69 differentially expressed FAM-related genes between 502 HNSCC samples and 44 normal samples from The Cancer Genome Atlas (TCGA) database. The HNSCC samples were divided into 2 clusters according to 69 differe… Show more

“…For instance, NUTF2 has been reported to be highly expressed in HNSCC, associated with a poor prognosis and related to immune cells, which may serve as a potential biomarker and target for HNSCC ( 56 58 ). Additionally, previous studies have established a 12-gene signature based on the fatty acid metabolism to predict the prognosis of HNSCC, which contain the long non-coding RNA, LINC00460, indicating its predictive role for HNSCC ( 36 , 59 , 60 ). Also, another long non-coding RNA, LINC02454, is linked to predicting the prognosis of thyroid cancer ( 61 , 62 ).…”

“…Of course, there existed some limitations in this study. Similar to many publications (49)(50)(51)(52), we only extracted data from public databases and did not validate these data with animal experiments and clinical specimens. In addition, the nomogram lacked imaging data, which are also significant for assessing the prognosis of HCC.…”

“…According to the previous studies, 15,20 All subjects were divided into high-and low-risk groups according to the median risk scores after each sample's risk score was calculated with the formula. To confirm the value of the above risk score model to predict patient prognosis, the log-rank test and Kaplan-Meier survival curves were used to compare the overall survival (OS) of the patients in both groups.…”

“…Consequently, tumor cells exhibit notable variations in protein, fatty acid, and glucose metabolism from normal cells 13 . Tumor cells are revealed to be able to more readily utilize the higher energy provided by fatty acid metabolism (FAM) 14,15 . Genes in the fatty acid metabolic pathway are known as fatty acid metabolism‐associated genes (FAMGs), whose reprogramming is an important guarantee for the continuous growth, proliferation, and migration of cancer cells 16,17 .…”
