Background. Monogenic scab resistance is an important trait of apple, useful to plant breeders. DNA markers provide a possibility to differentiate apple cultivars according to individual resistance determinants with high reliability and identify promising genotypes. The present study shows the results of the molecular genetic analysis of apple varieties, targeted at the Rvi2, Rvi4, Rvi6 and Rvi8 monogenic scab resistance loci.Materials and methods. Biological material was represented by apple cultivars of different environmental and geographical origin. Total genomic DNA was extracted from fresh leaves using CTAB methods according to the DArT protocols. The Rvi6 gene was identified with two markers, VfC (STS) and AL07 (SCAR). The presence of the Rvi4 gene was detected with the multiallelic SCAR marker AD13. The Rvi2 and Rvi8 genes were diagnosed with the SCAR marker OPL19.Results and conclusion. The Rvi6 gene was identified in 54.4% of genotypes, of which 91.9% were heterozygous, and 8.1% (cvs. ‘Svezhest’, ‘Freedom’ and ‘GoldRush’) homozygous dominant for this locus. The marker AD13-SCAR was detected in 25.0% of the studied forms (the putative genotype for the resistance gene is Rvi4Rvi4 or Rvi4rvi4). The marker OPL19-SCAR (Rvi2 and Rvi8 genes) was present in 73.5% of the analyzed forms. At least one of the studied molecular markers was present in the genome of 86.8% of genotypes. The appletree cultivars ‘Kandil Orlovsky’, ‘Krasulya’, ‘Sozvezdiye’, ‘Galarina’, ‘Priam’, ‘Redfree’ and ‘Witos’ are characterized by the combination of markers VfC, AL07-SCAR, AD13-SCAR and OPL19-SCAR in one genotype (the putative genotype for the resistance genes is Rvi2(Rvi8)Rvi4Rvi6rvi6). Cvs. ‘Antonovka zimnyaya’, ‘Antonovka krasnaya’, ‘Berkutovskoye’, ‘Geyzer’, ‘Pamyati Nesterova’, ‘Renet Simirenko’, ‘Terentyevka’, ‘Golden Delicious’, and ‘Telemon’ presumably have a homozygous recessive genotype for the studied resistance loci.

Genetic diversity and nucleotide sequence analysis of powdery mildew marker and Vf2RAD resistant gene in apple (Malus domestica) land races

Introduction: DNA sequencing-based methods and nucleotide sequence analysis have become the most common molecular approaches currently used for molecular typing purposes and phylogenetic diversity analysis. Methods: In this study, the nucleotide sequence variations of Powdery mildew resistance gene marker (CH03c02) and the apple scab resistance gene (Vf2RAD) beside phylogenetic diversity of seven apple landraces have been investigated. The two-locus have been successfully cloned and their nucleotide sequences were determined across all studied landraces. Results: Results of sequence alignment of the Powdery mildew resistant locus (CH03c02), compared with that of the published sequence of the same locus of Discovery genotype (HiDRAS), revealed that the nucleotide variations of this locus ranged from 1 to 28 nucleotide substitutions across all seven apple landraces. Whilst, the nucleotide variations of VF2RAD ranged from 2-8 nucleotide substitutions across all the investigated landraces. The highest genetic distance (0.062) was between Amara and Barwari. Whereas, the lowest genetic distance (0.0015) was found between each of the Lubnani, Rechard, Ispartal, and the Ahmadagha. The nucleotide sequences of the two loci were concatenated and implemented to build a Neighbor-Joining tree. The seven apple landraces were successfully grouped into two main genetic clusters (C1 and C2) in the phylogenetic tree. Conclusions: It can be concluded that the cloning approach used in the current study was found to be very successful and helpful for obtaining the full nucleotide sequences of these two loci. The investigated loci were displayed nucleotide variations among the studied landraces. And, finding of these variations was allowed the distinguishing and discrimination of these landraces.

Stripe rust, caused by Puccinia striiformis f. sp. tritici (Pst), is one of the most important diseases affecting wheat. In this study, seven gene-linked markers were used to identify the presence of stripe rust resistant genes in 51 accessions of synthetic hexaploid of wheat which were found to be resistant at seedling plant stage. Molecular marker-based gene identification showed the presence of Yr5, Yr10 and Yr15 in three accessions, Yr36 in three accessions, Yr48 in seven accessions, YrR61 in four accessions, and YrTP1 in ten accessions of resistant hexaploid of wheat. These gene-linked markers were also used for the detection of genetic diversity. A total of 68 alleles were detected by these seven gene-linked markers. The mean number of allele was 11.3 alleles per locus. Genetic diversity values ranged from 0.34 to 0.93, with highest genetic diversity value of 0.93 detected for marker Xwm477. The lowest genetic diversity value was observed for marker Xbarc167. The polymorphic information content value ranged from 0.33 to 0.92 with an average of 0.54. The highest number of alleles (n= 24) were detected for marker Xwmc477. The evidence in this study on the basis of genetic diversity and presence of Yr genes in synthetic hexaploid wheat accessions will be useful in further breeding programmes.
